Study On Quality Standard Of Radix Tetrastigmae

Radix tetrastigmae is a commonly used Traditional Chinese Medicine,has a long history in medicinal effect.And the flavonoids compounds of Radix tetrastigmae has been studied,with widely biological and pharmacological value.But the lack of quality standard and the disorded application seriously restrict the development and application of Radix tetrastigma.To provide theoretical foundation for the research of quality standard of Radix tetrastigmae,through establishing systematic quality evaluation methods,like identification（thin layer chromatography TLC identified）,check（water,total ash）,extract（alcohol-soluble and water-soluble extract determined）,and the content determination and so on.All the methods are according to the china pharmacopeia（2010 edition appendixⅡ,appendixⅥ,appendixⅨand appendixⅩ）.First of all,22 batch of Radix tetrastigmae water content of the sample top2.47%,the lowest 1.83%;the total ash content in the top 95.48%,the lowest65.17%;water-soluble extract the highest content 12.19%,the lowest 8.17%;alcohol soluble extract the highest content 15.69%,the lowest 10.87%.Secondly,Radix tetrastigmae thin layer of chromatography identify research shows that,the TLC spots clearly and with reference substance in the corresponding position,show the same color of the spots.NaNO₂-Al（NO₃）₃ colorimetric method was used for determination of total flavonoids in Radix tetrastigmae,and the method was simple and rapid.The content ranges of the flavonoids components were as follow:3.02⁶.02mg/g.Meanwhile,we established a new method of quality evaluation,quantitative analysis of multi-components by single marker,for the simultaneous determination content of resveratrol glucoside and resveratrol,and the methodology also was studied.By the content determination of 15 batches of different origin of medicinal materials,the content ranges of resveratrol glycoside and resveratrol were as follow:4.10⁷5.79(10^-3mg/g)and 3.10⁶.72(10^-3mg/g).To provide a scientific basis for quality control of different areas Tetrastigma hemsleyanum by establishing a HPLC characteristic fingerprint.The common pattern of HPLC characteristic chromatographic profile was established.There were 14 common peaks,two of which were identified in the pattern.The similarities of 10 batches of Tetrastigma hemsleyanum were evaluated,and all of them were greater than 0.900.This analysis method of HPLC characteristic chromatographic fingerprint is simple and reproducible,which it can provide a scientific basis for identification.Finally,extraction is investigated by testing scavenging effects of DPPH and determine the minimum in vitro inhibitory concentration（MIC）and diameter of inhibition zone against Staphylococcus aureus,Escherichia coli.The results showed that Tetrastigma hemsleyanum of extract exhibites a strong ability to remove DPPH,and the antibacterial effect is not so well.The Tetrastigma hemsleyanum all aspects datas of quality control system are obtained through this research.And the Tetrastigma hemsleyanum standard of quality was drafted on this basis,which provide a basis for quality assessment of medicinal materials.
