The Tetrastigma hemsleyanum Diels et Gilg(T.hemsleyanum)is a key cultivar of the new "Zhe-ba-wei".It is a traditional Chinese medicine for clearing away heat and detoxifying.It has not been included in the Chinese Pharmacopoeia.The "Zhejiang Traditional Chinese Medicine Processing Regulations(2015)" only has alcohol-soluble extracts in terms of quantitative detection indicators.Without any other quantitative control indicators of medicinal ingredients,it is impossible to carry out accurate,in-depth and systematic quality evaluation and quality control of different T.hemsleyanum.In view of this,this research intends to establish a more accurate and systematic multi-dimensional fingerprint and saccharide mapping detection method of T.hemsleyanum,and conduct quality evaluation research on T.hemsleyanum medicinal materials from different origins,different processing methods,and different years,which can improve more accurate quality evaluation methods and technical basis for exploring the quality formation process of medicinal materials,selecting suitable production areas and reasonable processing..The main conclusions are as follows:(1)To establish the HPLC-DAD-ELSD fingerprints and the polysaccharide fingerprints of T.hemsleyanum.The methods were be stable and reliable and can be used for quality evaluation research.(2)The content of active ingredients in T.hemsleyanum from 10 different producing areas was determined.It was found that the Duyun has the highest content of total flavonoids and polyphenols,which respectively were 37.909 mg/g,11.251 mg/g.The content of Suichang were second,which were32.025 mg/g,10.275 mg/g.The highest total polysaccharide was Suichang.Comparihg the wild and artificial planting,it showed that S2 has a high content of comprehensive active ingredients and the quality was closest to wild samples.Through multi-dimensional fingerprints and chemometric analysis,it can be seen that the content of T.hemsleyanum from different origins was quite different.Duyun and Suichang samples of T.hemsleyanum medicinal materials have higher comprehensive scores of principal component factors,and the quality is better than other origins.The content of monosaccharides in the polysaccharides was determined,and all the trilobite polysaccharides contained 8 monosaccharides,which was mannose,glucosamine,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose and arabinose.Among them,mannose,glucose,galactose and arabinose had the highest content,which were the main components in the polysaccharides of T.hemsleyanum.Analysis with the content of glucose,galactose and arabinose as indicators shows that the overall content of Suichang is higher than other origins.(3)Through the analysis of the active ingredient content of different processing methods,the multi-dimensional fingerprint spectrum and the polysaccharide sugar profile,it can be seen that the highest comprehensive content of fresh products was 33.919 mg/g,9.641 mg/g,and 37.543 mg/g.The active ingredient content in the freeze-dried is closest to the fresh.The principal component analysis shows that the comprehensive score of fresh products is the highest.The freeze-dried is second only to fresh products.Besides,fresh and freeze-dried monosaccharides have the highest content.(4)Determing the content of total flavonoids,total polyphenols and total polysaccharides in different ages,it was found that the content of active ingredients in artificially cultivated samples were increased with the years,and the content of fourth yearws was the highest.By comparing the multi-dimensional fingerprint patterns of different ages,it can be found that the quantity and content of 4 years is the best in all years,which are 37.503 mg/g,12.629 mg/g and 45.653 mg/g respectively.The principal component analysis shows that the 4 years sample has the highest comprehensive score of principal component factor,which means the best quality.After measuring the monosaccharide content in the polysaccharide,it can be seen that the overall content reaches the highest at the fourth year.In summary,the multi-dimensional fingerprints and polysaccharide fingerprints,which were established in this research,complete the quality evaluation standard of T.hemsleyanum.The best production area is S2,the best growth period is 4 years and the best processing method is freeze drying. |
