| Objective:Examined the effect of Tongxie Yaofang in different bowel movements:mice in different bowel movements and experiment on rat intestine movement and to explore the function of Tongxie Yaofang to IBS rat intestinal movement.The expressive effects of the Tongxie Yaofang to the ghrelin content that contented in the hypothalamus,spinal cord and colon tissue of IBS model.Dabated the relationship of IBS bowel movement and brain-gut axis control on the function of Tongxie Yaofang,and revealed the treatment mechanism of Tongxie Yaofang to IBS.Methods:Rhein-induced diarrhea model in mice,compound diphenoxylate-induced constipation model mice and normal mice were used to observe the effects of different dose of Tongxie Yaofang to the bowel movement of mice in each group,and through the effects on the small intestinal propulsion hyperthyroidism and normal mice intestinal propulsion movement of neostigmine-induced mice were used to observe the regulation effect of that Tongxie Yaofang to the movement of mice intestinal propulsive.Cleaning grade newborn Wister rats were choosed to screen the model through the AWR method,and the model were replicated with the methods of rectal stimulation combined with the expansion of maternal separation.Vitro methods were used to observe the effects of the Tongxie Yaofang to the IBS rat intestinal.The immunohistochemical was used to observe the effects of the Tongxie Yaofang to the content of the ghrelin 5-HT,Substance P in IBS rat hypothalamus,spinal cord and colon tissueResults:Defecation results in mice show that Tongxie Yaofang can both ease the diarrhea caused by rhubarb(P<0.05,P<0.01)and promote the diphenoxylate-induced bowel constipation model mice,a low dose has no significant effect on the normal,while normal dose can promote bowel movement mice(P<0.01).The study of small intestine carbon of mice show that Tongxie Yao.fang can order against the neostigmine intestinal propulsive caused by neostigmine-induced mice(P<0.05),and had no effect on normal mice.The study of the effects on Tongxie Yaofang to IBS rat intestinal movement show that,Tongxie Yaofang can reduce the isolated peristaltic contraction tension and frequency on IBS rat model(P<0.05).It has a high expression of the 5-HT and Sp in IBS model rats hypothalamus,colon and spinal cord,Tongxie Yaofang can lower the 5-HT and Sp in IBS modcl rats hypothalamus,colon and spinal cord significantly(P<0.01)Conclusions:Tongxie Yaofang has regulatory effect on different gastrointestinal movement motion state on mice and IBS rat,it can also reduce the expression levels of 5-HT and SP in IBS model rats hypothalamus,colon and spinal cord,suggesting that the Tongxie Yaofang’s regulate function to bowel movement has relation to regulate the content of the brain-gut peptide in brain-gut axis. |
