Background and objectives:Ovarian cancer(OC)is the highest mortality of female genital malignant tumor.Immunologic escaping is one of the main factors leading to the poor prognosis of OC patients.Indoleamine-2,3 dioxygenase(IDO),is the key factor which mediates the immunodepression and tumor immunologic escaping in ovarian carcinoma.Inhibiting the expression or the activity of IDO can improve the immunity of ovarian cancer patients,and ameliorate the prognosis.Berberine can inhibit the activity of IDO,but if berberine can down regulated the expression of IDO and aryl hydrocarbon receptor(AhR)is lack of research.This experiment will study the effects of berberine on the expression of IDO,AhR mRNA and the activity of IDO in human ovarian cancer cells.Methods:Testing the inhibitory effects of berberine on human ovarian cancer cells SKOV-3 and A2780by crystal violet method;Using RT-PCR to detect SKOV-3 and A2780 cells,IDO,AHR mRNA expression after the effects of berberine with different concentrations at different time;Colorimetric assay was used to detect the changes of the dosing of tryptophan kynurenine to IDO in the supernatant of the cells before and after the effect of berberine.Results:1、Our study found that A2780 does not have high expression of IDO,and the high expression of IDO may be directly related to myd88;2、Berberine can inhibite the growth of SKOV-3 and A2780 cells in vitro(IC500 are2.48±0.269μmol/L and 2.41±0.157μmol/L,respectively);3、Berberine could significantly inhibit the expression of IDO and AHR mRNA in the SKOV-3cells,and the inhibitory activity increasing with drug concentration and the time enhancement,while berberine concentration dependently reduced SKOV-3 cell supernatant of tryptophan depletion and kynurenine production;4、Berberine could significantly inhibit the expression of AHR mRNA in the A2780 cells without high expression of IDO,and decrease the concentration of tryptophan in supernatant of A2780 cells and the production of kynurenine.Conclusion:1、Our study found that A2780 is not high expression of IDO,the there is not related reported in existing literature.2、The effects of berberine on the expression of AHR mRNA and the depletion of tryptophan in ovarian cancer cells is not wholly linked to the high expression of IDO.3、Berberine can overcome the immunodepression of ovarian cancer,and maybe,could well prove effective in both of type I and type II ovarian cancer.
