Objective:1.To clarify the relationship between signal transduction and activator of transcription Stat5 and pruritus;2.To identify the relationship between the effect of acupuncture on relieving itch and Stat5;Methods:Twenty-four,8-10 weeks old,weighing 20-25 g,Stat5ff mice and Stat5 ffcre mice were randomly divided into Stat5 ff Histamine group,Stat5 ff Compound48/80 group,Stat5 ff chloroquine group,Stat5 ffcre histamine group,Stat5 ffcre Compound48/80,and Stat5 ffcre Chloroquine group(n=4/per group).After subcutaneously injection of3 types of allergenic substances,Histamine,Compound 48/80,and chloroquine,the number of scratching was recorded every 10 minutes and the observation time was continued for 30 minutes.The healthy Babl/C mice,male,10 weeks old,weighing 20±2g,were randomly divided into control group,saline group,histamine model group,histamine+2Hz electroacupuncture group,2Hz electroacupuncture group and sham acupuncture group(n=8/per group).Electroacupuncture was adminstered to the bilateral Zusanli acupoints.After the acupuncture treatment,20 ul saline or 50 ug histamine was injected into the subcutaneous of neck and back.The number of scratches within 30 minutes was recorded.Western Blot was used to detect the expression of STAT5 in DRG,hypothalamus and hippocampus.Results:After subcutaneous injection of histamine,Compound 48/80 and chloroquine,the scratching frequency of Stat5 ffcre mice was significantly lower than that of Stat5 ff mice within 30 minutes(P<0.01).There was no significant difference in the number of scratching times among the control group,saline group,and 2Hz electroacupuncture group(P>0.05).Compared with the control group,saline group and 2Hz electroacupuncture mice,the number of scratching in the histamine model group mice within 10 min and 10-20 min after administration were respectively increased,and the difference was statistically significant(P<0.01).The number of scratching in histamine+2Hz electroacupuncture group was significantly lower that in histamine model group(P<0.01).There was no significant difference in the number of scratching mice in the sham acupuncture group compared with histamine group(P>0.05).No statistically significant difference in the expression of STAT5 in DRG,hypothalamus and hippocampus prestented in this study(P>0.05).Conclusions1.Stat5 conditional knockout mice have a significantly reduced scratching bouts in different acute pruritus condition.2.Low frequency electroacupuncture(2Hz)has good antipruritic effect on acute itch.3.Stat5 may not be involved in antipruritic effect of acupuncture.
