Objective:To observe the effects of estrogen on renal ischemia-reperfusion injury and T lymphocyte differentiation after RIRI.Methods:Eighty female Sprague-Dawley rats were treated with ovariectomy before establishing renal ischemia-reperfusion injury model.The rats was divided into four groups:Normal group,Sham group,I/R group and E2+I/R group.Hematoxylin-eosin(HE)staining was used to observe renal tubular lesions(paller’s score).An automatic biochemical analyzer was used to detect the level of blood urea nitrogen(BUN)and creatinine(Cr).Flow cytometry was used to detect changes of CD3,CD3+CD4+,CD3+CD8+,CD4+IFN-γ,and CD4+IL-4 cells in peripheral blood of each group.ELISA was used to detect levels of IFN-γand IL-4 in plasma and renal tissue.Results:Compared with Sham group,the degree of tubular injury increased;levels of BUN,Cr,CD3,CD4,CD4+IFN-γcells and IFN-γall increased in I/R group(P<0.05).The number of CD8,CD4+IL-4 cells and the expression of IL-4 decreased(P<0.05).Compared with I/R group,tubular injury in E2+I/R group decreased,and the levels of BUN,Cr,CD3,CD4,CD4+IFN-γcells and IFN-γexpression all decreased(P<0.05).The content of CD8,CD4+increased and the expression of IL-4increased(P<0.05).Conclusion:Estrogen may protect the kidney from renal ischemia-reperfusion injury by affecting the differentiation of T lymphocytes and the release of inflammatory factors.
