| Objective: To investigate whether estrogen(Estrogen,E2)can improve the systolic and diastolic function of renal arteries after renal ischemia-reperfusion injury by regulating gap junctions,and to further explore the possible mechanism of estrogen on protective effects of renal ischemia-reperfusion injury.Methods:(1)Eight healthy male SD rats were randomly divided into control group(n=20),sham operation group(n=20),renal ischemia-reperfusion injury group(n=20),estrogen intervention group(n=20).Control group: do not do any treatment.Sham operation group: resection of the right kidney,but not the left side of the renal pedicle,45 minutes after the closure of the abdominal cavity.Ischemia-reperfusion injury group: resection of the right kidney,with non-invasive artery clamp clip left kidney pedicle,continuous ischemia 45 min,restore blood flow.Estrogen intervention group: experimental animals were given estrogen(100 ug / kg)subcutaneously once a day for three days,and the rest of the steps were similar to those of ischemia-reperfusion injury.The levels of serum creatinine(Cr)and urea nitrogen(BUN)were measured by automatic blood biochemical analyzer.The histopathological changes of renal tissue were quantified by HE staining of renal tissue sections.(2)In the acute separation of the interosseous arteries of the rats,the 2-3 mm blood vessel was obtained.The contractile response of the renal interlobular arteries was induced by the non-endothelium-dependent vasoconstrictor KCl.And the effect of the gap-blocking inhibitor 2-APB on the vascular contractile response of the rat interosseous artery.(3)The expression and distribution of Cx43 on the interlobular arteries were observed by immunofluorescence.(4)QT-PCr and Western-blot were used to detect the expression of connexin in the interlobular artery of rats.Results:(1)Estrogen can significantly reduce the level of Cr and BUN in rats with ischemia-reperfusion injury and improve the degree of renal damage.Compared with the normal group,the levels of Cr and BUN were significantly increased in ischemia-reperfusion injury group,and Cr and BUN were significantly decreased after ischemia-reperfusion injury(P <0.05).HE showed ischemia-reperfusion injury A large number of inflammatory cells infiltration,proximal tubule epithelial cell vacuolar degeneration,necrosis,renal tubular lumen expansion,visible tube and necrotic exfoliated cells,peripheral vascular dilatation congestion,Paller score was significantly higher than the normal group and false(P <0.05),but the estrogen intervention group also showed damage of renal tissue structure,but the renal interstitial inflammatory cell was significantly reduced,Paller injury score was significantly lower than ischemia and reperfusion injury group(P <0.05).(2)estrogen on the contraction of blood vessels and gap junctions related to communication.The contraction rate and rate of interosseous arteries were significantly decreased after renal ischemia-reperfusion injury,and the rate and rate of interlobular artery contraction were significantly higher than that of ischemia Perfusion injury group,the difference was statistically significant(P <0.05).(3)Estrogen can reduce the expression and distribution of Cx43 on the interosseous artery smooth muscle of renal ischemia-reperfusion injury.The expression of Cx43 on renal smooth muscle was significantly enhanced after renal ischemia-reperfusion injury,and the expression of Cx43 was significantly decreased after estrogen intervention.(4)Estrogen can decrease the expression of Cx43mRNA and Cx43 in the interlobular arteries after renal ischemia-reperfusion injury.The expression of Cx43 m RNA and Cx43 in the interosseous artery of renal ischemia-reperfusion injury was significantly higher than that in the normal group(P <0.05).The expression of Cx43 m RNA and Cx43 was significantly lower than that of the ischemia-reperfusion injury group(P <0.05).Conclusion: Cx43 plays an important role in the regulation of systolic and diastolic function of rat interosseous artery.The expression of Cx43 on the smooth muscle cells of the renal interlobular artery after ischemia-reperfusion injury is inhibited by estrogen,and the GJ function is reduced,Promote vasodilatation,effectively reduce renal ischemia and reperfusion injury. |
