Objective:By establishing a mouse model of adenomyosis,we observed the effects of NYKFP,which has the effect of removing dampness and removing blood stasis,on the expression of ras,COX2,PGE2 and P450arom in different estrous cycle and ectopic endometrium of mice,in order to explore the mechanism of NYKFP in the treatment of adenomyosis.This laid a foundation for clinical medicine differentiation.Methods:A total of 216 ICR female mice without gestation at 8 weeks of age were transplanted into the uterus to make adenomyosis mice.Then the model mice were randomly divided into 3-month and 6-month model groups,NYG,NYZ,NYD group,DE group and YSXT group,plus Sham group and Normal group,24 in each group.The Normal group,Sham group and 3-month model group were fed with respectively,3 months later,the uterus was sampled in each group to determine whether the mouse model of adenomyosis was successfully established.After the successful establishment of the model,the other groups were given intragastric administration of drugs.After3 months,to sample the uterus.The pathological sections of uterine tissue were stained with HE before and after modeling and administration of drugs in mice.The degree of infiltration of the glands and tissues of the uterus and grading scores under the microscope;and the immunohistochemical staining was used to detect the difference of expression of ras,COX2,PGE2 and P450arom in different cycles on eutopic and ectopic endometrium.Results:1.There was no significant difference between Sham group and Normal group(P >0.05).2.After modeling,the pathological grading of the model group in 3-month and 6-month was significantly higher than that in the Normal group(P< 0.01),and the pathological grade in the 6-month model group was significantly higher than that in the 3-month group(P< 0.01).3.After modeling,the expression of ras,COX2,PGE2,and P450 arom in the eutopic and ectopic endometrium in the 3-month and 6-month group was significantly higher than that in the Normal group during estrus and interoestrus period(P< 0.01),and the expression levels in 6-month model group were significantly higher than those in the 3-month model group(P< 0.01).4.After modeling,the expression of ras and COX2,PGE2,P450 arom in ectopic endometrium during estrus and interoestrus was significantly higher than that in eutopic endometrium(P< 0.01).5.After administration,compared with the 6-month model group,the expression of ras and COX2,PGE2,P450 arom on NYG,NYZ,DE and YSXT was were significantly lower than that of6-month model group(P< 0.01),and their expression of NYG was significantly lower than that of DE group and YSXT group(P< 0.01).And the difference was statistically significant in different dose groups(P < 0.01).6.Correlation analysis showed that there was a positive correlation between ras and COX2-PGE2-P450arom(P< 0.01).Conclusions:1.The occurrence of adenomyosis was related to the high expression of ras and COX2-PGE2-P450 arom,and there was a positive correlation between them.2.The curative effect of high dose NYKFP was better than that of that of Dan’e Fukang decoction and pregnancy trienone,and the effect of high dose NYKFP was better than that of medium and low dose.3.The treatment of adenomyosis with NYKFP may be achieved by reducing the level of ras,COX2,PGE2,P450 arom to weakening the estrogen effect.4.Mechanical injury during operation will not affect the results of experimental modeling. |
