| Objective: By comparing the amount of typeⅡcollagen among the autologous 6th,7th,and 8th costal cartilages,to provide theoretical basis of autologous cartilage transplantation at the molecular level.Methods: From 8 to 12 years old patients performed ear reconstruction surgery with autologous costal cartilage,collecting every 20 costal cartilage tissue samples of 6th rib,7th rib,and 8th rib.Dividing into three groups according by different sources of parts.Each group contents 20 samples,and 60 samples total.The level of m RNA of typeⅡcollagen in the 6th,7th,8th costal cartilages in patients were tested by florescent real-time quantitative PCR.The protein content of typeⅡcollagen and the amount of chondrocytes in the autologous 6th,7th,8th costal cartilage in patients were detected by immunohistochemistry and HE staining.Results: 1)there is no significantly difference of the level of type Ⅱ collagen m RNA among 6th,7th,and 8th costal cartilages(9.3 ± 1.2,8.98 ± 0.5,9.36 ± 1.3);2)there is no significantly difference of the relative gray value of type Ⅱ collagen in 6th,7th,and 8th costal cartilages(14517.8 ± 398,15368.3 ± 736,13992.05 ± 802);3)there is no significantly difference of the amount of chondrocytes in 6th,7th,and 8th costal cartilages.Conclusion: Through this study found out that there is no significant difference in the comparison of m RNA expression and relative gray values in type Ⅱ collagen in 6th,7th and 8th rib.Also,there is no significant difference in the comparison of costal cartilage cell count in in 6th,7th and 8th rib.This result can be concluded that during autologous cartilage transplantation,which considerated of selecting dawning autologous cartilage tissue parts at the molecular level,is no significant difference in 6th,7th or 8th rib. |
