Objective: Helicobacter suis is capable of infecting all kinds of animals including humans,and H.suis infections can lead to gastric mucosa-associated tissue(MALT)lymphoma.Gastric Mucosa associated lymphoid tissue lymphoma is a variety of factors involved in the development of progressive disease,a large number of studies have shown that obesity is a chronic low-grade inflammatory state,believing that obese are factors may be involved in the formation process of lymphoma gastric Mucosa associated lymphoid tissue(Mucosa associated lymphoid tissue MALT).Recently,the study showed that interleukins-22(IL-22)expressed higher in H.pylori-infected mice,positively correlated with the virulence of H.pylori,and the physical and chemical property of H.suis is similar to H.pylori,both of them belong to gram-negative bacillus,and has been reported to associate with gastritis,peptic ulcers,and gastric cancer.Furthermore,gastric mucosa-associated lymphoid tissue lymphoma occurs more frequently in H.suis-infected mice compared with H.pylori-infected mice and it can form in almost 100% of C57BL/6mice at 6 months after infection,suggesting that H.suis has a greater tendency to induce gastric MALT lymphoma.Through the study of H.suis infecti on in obese mice,further explore the expression and significance of IL-22 in H.suis infection of obese mice,in order to further clarify the obese factor promote the development of tumor mechanism,then provides a new research direction and theoretical basis.Methods: 1.Establishment of obesity mice model with H.suis infection and the express of IL-22 、 IL-22R1 and the chemokines CXCL13 : Forty female C57BL/6 mice were randomly divided into four groups(ten mice per group),which are named as normal control(NC)group,high fat diet(HFD)group,H.suis infection(HS)group and H.suis infection+ high fat diet(HS+HFD)group.The mice of NC group and HFD group were given ordinary diet,and the mice of HFD group and HS+HFD group were given high fat diet from the third week postinfection.After 24 weeks different diet,all of the mice were sacrificed to collect gastric tissues.The number and size of gastric lymphoid follicles were detected by HE staining and the m RNA expression levels of IL-22 and its receptor IL-22R1,chemokines CXCL13 in the gastric mucosa of mice were detected by real-time PCR.Results: 1.The weight of HFD group was significantly higher than that in the NC group,the weight of HS+HFD group was significantly higher than that in the HS group.There was no significant difference in weight between NC and HS group,HFD and HFD+HS group,indicating that H.suis did not significantly affect the weight change of mice.2.H.suis infection may induce the formation of gastric mucosal lymphoid follicle in mice;Compared with the HS group,the number and size of gastric mucosal lymphatic follicle in the HFD group increased significantly,and the m RNA expression level of the gastric mucosa IL-22 and CXCL13 increased significantly.3.The expression of IL-22 R1 in obese mice was decreased,which are consistent with the results of the study abroad,the study suggests that the expression of IL-22 R1 in obese mic was lower than the normal mice,and IL-22 R1 defects from the defective mice prone to weight gain and risk of insulin resistance.Conclusion: This study successfully established the obesity mouse model of H.suis infection.2.Expression of IL-22 was increased in obese mice with high fat diet,and high fat diet downregulate the expression of IL-22R1.3.H.suis infection increased expression of IL-22,ILl-22R1 and CXCL13 in mice,especially increased the expression of IL-22.4.H.suis could induce the formation of gastric mucosal lymphatic follicle in mice,but it is more likely to induce the formation of gastric mucosal lymphoid follicle in obese mice. |
