Objective: The aim of this study was to compare plasma secreted frizzled-related protein 5(SFRP5)levels between subjects with different glucose tolerance status,and subjects with the same glucose tolerance status but different obesity degrees,analyze the correlations between plasma SFRP5 level and gender,age,obesity,blood pressure,glucose and lipid metabolism,insulin resistance,β cell function,and inflammatory,and further investigate the independent correlated factors of plasma SFRP5.Methods: 354 subjects were recruited from outpatients in the special medical department of Beijing Jiangong Hospital from January 2017 to December 2017.General information of every subject was collected,and oral glucose tolerance tests(OGTT)were performed.According to the OGTT results,all subjects were divided into three groups: the normal glucose tolerance(NGT)group containing 132 subjects,the impaired glucose regulation(IGR)group containing 107 subjects and the newly diagnosed type 2 diabetes mellitus(T2DM)group containing 115 subjects.Biochemical indicators and plasma SFRP5 were measured.The levels of plasma SFRP5 among three groups were compared using ANOVA.Further,each group was divided into abdominal obesity subgroup and normal waist circumference subgroup according to the genders and waist circumferences.The levels of plasma SFRP5 between the two subgroups of each glucose tolerance status were compared using Student’s t test.The correlations between plasma SFRP5 levels and gender,age,waist circumference,body mass index,blood pressure,fasting plasma glucose(FPG),2h plasma glucose(2hPG),glycosylated hemoglobin(HbA1c),fasting insulin(FIns),fasting C-peptide(FCP),total cholesterol,triglyceride,high-density lipoprotein cholesterol,low-density lipoprotein cholesterol,high-sensitivity C-reactive protein(hsCRP)levels,homeostasis model assessment of insulin resistance(HOMA-IR),and homeostasis model assessment of β-cell function(HOMA-β)were analyzed.Multiple linear stepwise regression was used to analyze the independent correlated factors of plasma SFRP5.Results:1.There were no significant differences between gender,age and obesity degrees among different glucose tolerance status groups.The plasma SFRP5 level of the IGR group was significantly lower than that of the NGT group(P<0.05),and the plasma SFRP5 level of the T2 DM group was significantly lower than that of the IGR group(P<0.01).2.In each group with the same glucose tolerance status,while no significant differences were found in gender,age or plasma glucose levels,the plasma SFRP5 level of the abdominal obesity subgroup was significantly lower than that of the normal waist circumference subgroup(P<0.05 between subgroups of the NGT group,P<0.01 between subgroups of the IGR group and the T2 DM group).3.The plasma SFRP5 was negatively correlated with HbA1 c,FPG,2hPG,FIns,FCP,HOMA-IR(all P<0.01),but not correlated with HOMA-β.Plasma SFRP5 was negatively correlated with hsCRP and waist circumference(both P<0.01).4.Multiple linear regression analysis showed that HbA1 c and FIns were the key influence factors of plasma SFRP5(P<0.01).Conclusions: Plasma SFRP5 is negatively related to abdominal obesity,insulin resistance and inflammation degree.The reduction of SFRP5 levels may be related to T2 DM,especially T2 DM accompanied by abdominal obesity.
