Objective:To investigate secreted frizzled-related protein5(SFRP5)in T2DMpatients and obese subjects and explore the correlation of serum SFRP5level with otheranthropometric and biochemical parameters.Methods:Forty-eight newly diagnosed patients with type2diabetes mellitus(T2DM)and forty subjects with normal glucose tolerance(NGT)were enrolled and then each groupwas divided into normal weight(NW)and overweight/obesity(OW/OB)subgroups,according to18.5kg/㎡<BMI<25kg/㎡or BMI≥25kg/㎡. Fasting serum SFRP5levelswere measured using ELISA. We also determined other anthropometric and biochemicalparameters in all subjects.All data were analyzed by SPSS17.0.Comparion between twogroups was analyzed using t-test while analysis of variance(ANOVA)was used for foursubgroups comparison. Correlation analysis and multiple linear stepwise regressionanalysis were used to perform the interrelationships between SFRP5and otheranthropometric and biochemical parameters. The difference was considered statisticallysignificant when the two-tailed P value less than0.05.Results:The1evel of serum SFRP5in T2DM group was significantly lower than inNGT group(104.30±32.32pg/mL vs.150.32±27.87pg/mL,P<0.01).Compared with NWsubjects or NW subgroup in each group,serum SFRP5level in OW/OB subjects orOW/OB subgroup was significantly decreased.Partial correlation analysis showed thatserum SFRP5level negatively correlated with BMI、WC、WHR、SBP、DBP、FPG、HbA1c、hsCRP、TC、IL-6、TNF-α、FINS、HOMA-IR(r=-0.400,-0.436,-0.461,-0.217,-0.350,-0.510,-0.491,-0.400,-0.395,-0.273,-0.471,-0.427,-0.532,P<0.05or P<0.01),whereas positively with HDL-C(r=0.609,P<0.01).Furthermore,multiple linear stepwise regression analysis indicated that HDL-C, HOMA-IR andTNF-α were independent factors influencing serum SFRP5level(β=40.54,-6.998and -0.221respectively,P<0.05).Conclusion:Serum levels of SFRP5were significantly reduced in T2DM and obesesubjects. It may become a new way of predicting,preventing,diagnosing and treatingT2DM and obesity.
