| Objective With the large-scale cultivation of Chinese medicinal materials and the continuous expansion of cultivation areas,the quality of medicinal materials has changed to a certain extent.In order to ensure the superior quality,value and clinical efficacy of medicinal materials,it has become an inevitable requirement to improve and perfect the commodity specification standards.This study took Angelica sinensis of Gansu as the research object and systematically studied its commodity specifications and grades from both the sensory and chemical evaluations which proposed a reasonable and scientific method for the classification of commodity specifications and grades of Angelica sinensis.Methods To collect Angelica sinensis from different producing areas in Gansu Province,the initial classification was carried out according to the existing standards,then the appearance traits of medicinal materials with different specifications and grades were described and quantified,used microscopic and thin layer identification to identify medicinal materials of different specifications and grades qualitatively.At the same time,according to the general rules of the Chinese Pharmacopoeia(four parts,2015 edition),the safety of samples was determined.The content of active components in Angelica sinensis of different specifications and grades was determined,the HPLC fingerprint of Angelica sinensis with was established.UV method was used to determine the content of polysaccharides,and multivariate statistical methods were used to judge the correlation between commodity specifications and grades and investigation indexes.Results(1)The head diameter,main root diameter and single weight were determined by statistical analysis of sensory quantitative indexes.The microscopic identification of samples from different producing areas had some differences,its basic composition and structure were similar,while in the same place,there is no obvious difference among different commodity grades.The number,size and color depth of the spots in each place of production were slightly different in the identification of thin layer,which indicated that the species and contents of active component in different cultivation areas were different.However the number and color of spots among different commodity grades in the same origin showed no significant difference.The contents of impurity,moisture,ash and extract of Angelica sinensis of different commodity specifications and grades were in accordance with the provisions of the Pharmacopoeia,and SO2 residue,heavy metals and harmful elements limit,pesticide residue were not exceeded the standard.(2)The content of chlorogenic acid in Angelica sinensis was 0.0199%0.1538%,ferulic acid content between 0.0648%0.1456%,Z-ligustilide content between 0.5207%1.8797%,and polysaccharide content between 1.3603%4.4770%.The HPLC fingerprint of Angelica sinensis in Gansu represented high similarity,and there is no significant difference among different commodity specifications and grades.(3)Through the use of statistical software for data correlation analysis,principal component analysis,cluster analysis,Fisher discriminant analysis,etc.,it was found that there was significantly negative correlation between chlorogenic acid and commodity grades,but leachate,ferulic acid,and Z-ligustilide were significantly positively correlated with the commodity grades.The order of correlation between sensory quantification index and the content of chemical components was single weight>head diameter>main root diameter.It can be seen that the original grade of Angelica sinensis based on the weight of the division has a certain rationality.Conclusion According to the research,three methods for the determination of the content of active components were accurate and reliable,which was of great crucial to evaluate the quality of different commodity specifications and grades,and the established HPLC fingerprint method was stable and feasible.It provides a reference for the commodity specifications and grades research and quality evaluation of Angelica sinensis,meanwhile perfects and revises that division of the commodity specifications and grades,and the classification method of Angelicae sinensis Radix in Gansu Province reflects the scientific connotation of"differentiation on quality". |
