Gentiana macrophylla Radix is a very important Chinese Materia Medica that was first recorded in Shen Nong Ben Cao Jing. As one of the traditional Chinese medicine, Gentiana macrophylla Radix has been widely used in the treatment of rheumatism, activating meridians to stop pain, asthenic fever and damp-heat disease. Its high medicinal value causes indiscriminate digging, further lead to greatly damage to wild resources. And it has been included in the List of National Key Protected Wild Medicinal Materials and is the third level of the key protection plants. There are other species, local supplies, could be used as Gentiana macrophylla Radix which alleviate the shortage of this kind of medicinal materials. However, the various and endangered species of wild Gentiana macrophylla Radix made it impossible to meet current market transactions if only use the standard of commercial specifications and grades of 76 Kinds of Chinese Herbal Medicinal.The market survey showed that the market transaction is chaos of Gentiana macrophylla Radix. Whether it is the specification level or the price, there is no uniform standard, and the traditional market evaluation standard is isolated from modern evaluation index of Gentiana macrophylla Radix. In this study, using the morphology description, macroscopical identification, microscopic character and TLC method to study four genuine crude drug species and local supplies of Gentiana macrophylla Radix, and develope the standard of commercial specifications and grades for collected 81 samples of Gentiana macrophylla Radix. And determine of 7 indicative constituents of Gentiana macrophylla Radix by UPLC. Though the typical correspondence analysis and Fisher discriminant analysis etc. to cluster the indicative constituents of Gentiana macrophylla Radix and develop the level of quality. Then, combined correlation analysis to selection the indexes of crude drugs quality, and classified comprehensive quantitative level for different commercial specifications and different grades of the same commercial specification of wild and cultivated Gentiana macrophylla Radix. The main study results as follows:1. There are great differences in morphology and organization among 6 medicinal plants species of Sect. Cruciata Gaudin which could been discerned by macroscopical identification, microscopic character and TLC method.2. The standard of commercial specifications and grades of Gentianae Macrophyllae Radix was developed as cultivated Luobojiao(first- class, second- class, third class), cultivated Mahuajiao(first- class, second- class), cultivated Xiaoqinjiao(first- class, secondclass), wild Luobojiao(first- class, second- class, third class), wild Mahuajiao(first- class, second- class), wild Xiaoqinjiao(first- class, second- class).3. There are great differences of the contents of 7 indicative constituents among 6 medicinal plants species of Sect. Cruciata Gaudin. And the higher of total loganic acid and gentiopicroside content was in Radix of Gentiana macrophylla, G. crassicaulis, G. officinalis and G. siphonantha; the second was in the Radix of G.straminea; the lowest was in the Radix of G. dahurica.4. Whether wild or cultivated, the quality of different commercial specifications of Gentiana macrophylla Radix was that Luobojiao was the best,Mahuajiao was average, and xiaoqinjiao was inferior.5. The quality of different commercial grades of Gentiana macrophylla Radix was that the quality of cultivated Luobojiao, the third class was the best, the second class was the second, first class was the worst, and there was a significant correlation; the quality of wild Luobojiao, the second class was the best, and the first class was the second; the quality of Mahuajiao,the first class was the best,and the second class was the second; the quality of Xiaoqinjiao, the second class was the best, and the first class was the second, but there was no significant correlation. |
