Hygrophorus lucorum kalchbrs,also known as Xiao huangmo,belong to Fungi,Basidiomycetes,Agaricales,Hygrophoraerae.It has been reported that the polysaccharide has many anti-tumor,and antioxidant activities.At present,the research on the polysaccharides of Hygrophorus lucorum kalchbrs is mainly focused on the extraction and bioactivity of polysaccharides.However,no reports have been found on the systematic classification and structural analysis of the polysaccharides.In order to understand the composition and structure of different kinds of polysaccharides in Hygrophorus lucorum kalchbrs,we designed the research contents of this subject:The specific results are as follows: the dried Hygrophorus lucorum kalchbrs are used as experimental materials.Using boiling water,precipitating with 65% ethanol alcohol,collecting sediments after drying,The total polysaccharide was obtained,and the yield was 7.1%(the total sugar content was 67.3%,the protein content was 5.0%,the ash content was 6.8%,almost no uronic acid and nucleic acid was found.The monosaccharide composition analysis shows that the crude polysaccharide was mainly composed of Glc(48.6%)and Gal(20.7%),In addition,it also contains Man(15.0%)and Fuc(8.1%),and a small amount of Ara(3.3%)and GlcA(2.5%).DEAE-Cellulose ion exchange chromatography is used to classify the charges of WHLP,The neutral fraction(WHLP-N)are obtained by distilled water elution and the acidic fraction(WHLP-A)eluted by 0.35 M NaCl.Further purification was carried out by using SepharoseCl-6B column chromatography to obtain final homogeneous fractions WHLP-N-b and WHLP-A-b.Molecular weight determination,monosaccharide composition analysis,methylation analysis and NMR analysis show that WHLP-N-b molecular weight is 12 kDa,which is the structure of α-D-1,6-Galp,the non-reducing ends Manp of the β-structure and Fucp of the α-structure are connected to the C-2 position of the Galp,and it is Fucomannogalactans.WHLP-A-b molecular weight is 3 kDa,which is the structure of β-1,6-D-Glcp,the non-reducing ends Glcp of the β-structure,β-1,3-D-Glcp,α-1,4-D-Manp,and α-1,2,6-D-Galp are connected to the O-3 position of the Glcp,the structure is.Mannogalactoglucans.In this paper,the systematic purification and the study of the structure of the polysaccharides from Hygrophorus lucorum kalchbrs,enrich the structural information of the polysaccharides and laid a foundation for exploring the relationship between the structure and the activity of the polysaccharides.At the same time,it also provides a theoretical basis for the opening of polysaccharide drugs.
