| Polygonum cuspidatum is a dry rhizome and root of Polygonum cuspidatum Sieb.Et Zucc.At present,there are few reports on polysaccharides from Polygonum cuspidatum,which are limited to the extraction process of polysaccharides from Polygonum cuspidatum and the activity of crude extract.The isolation and purification of Polygonum cuspidatum polysaccharide and its structural characteristics are not reported.The structure of Polygonum cuspidatum polysaccharide was studied in this experiment,and the α-glucosidase inhibitory activity of Polygonum cuspidatum polysaccharide was studied in this paper,which provided experimental basis for the comprehensive utilization of Polygonum cuspidatum.1、Three polysaccharides of Polygonum cuspidatum were prepared by ethanol precipitation,which were PPa,PPb and PPc,respectively.The yields were 0.53%,0.82% and 0.63%,respectively.PPa,PPb,PPc By DEAE-52 anion exchange,dextran gel,agarose gel column to obtain five components named PPa-Ⅰ-1、PPb-Ⅰ-1、PPb-Ⅰ-2、PPc-Ⅰ-1、PPb-Ⅱ-1.2、The content of polysaccharides and polysaccharides were determined by spectrophotometry.The results show: The contents of neutral sugar in PPa,PPb and PPc were 55.74%,59.50% and 17.15% respectively,and the contents of acid sugar were 4.26%,6.41% and 1.71% respectively,and the content of protein sugar was 19.41%,9.85% and 12.71% respectively.The contents of neutral sugars in PPa-Ⅰ-1,PPb-Ⅰ-1,PPb-Ⅰ-2,PPc-Ⅰ-1 and PPb-Ⅱ-1 were 99.85%,99.34%,99.65%,99.38%,83.47 PPb-Ⅰ-1,PPb-Ⅰ-1,PPb-Ⅰ-2,PPc-Ⅰ-1,does not contain acidic sugar,The acid sugar content of PPb-Ⅱ-1 was 12.83%.PPa-I-1,PPb-I-1,PPb-I-2,PPc-I-1 and PPb-II-1 did not contain protein.3、The composition of polysaccharides and homologous polysaccharides of Polygonum cuspidatum were analyzed by PMP-pre-column derivatization high performance liquid chromatography(HPLC).PPa is mainly composed of glucose,PPb is mainly composed of glucose,galactose and arabinose,The molar ratio is 35: 2: 1;PPc is mainly composed of glucose,galactose,arabinose and rhamnose,The molar ratio is 29:2:4:1;PPb-Ⅰ-1,PPb-Ⅰ-1,PPb-Ⅰ-2 and PPc-Ⅰ-1 are mainly composed of glucose composition content of 99%;PPb-Ⅱ-1 mainly consists of galactose,arabinose,rhamnose,glucose Uronic acid,the molar ratio 10:6:1:1.PPa-I-1,PPb-Ⅰ-1,PPb-I-2,PPc-I-1 and PPb-Ⅱ-1 were analyzed by high performance gel permeation chromatography.The results showed that there were single symmetric narrowing peaks,which were homogeneous polysaccharides with molecular weights of 2700 Da,7900 Da,5100 Da,2500 Da,49.20 KDa.4、Through the infrared spectrum,methylation combined with GC-MS,a variety of two-dimensional nuclear magnetic resonance spectroscopy and other means.PPa-I-1 repeating structural units are: PPb-Ⅰ-1 repeating structural units are: PPb-Ⅰ-2 repeating structural units are: PPc-Ⅰ-1 repeating structural units PPa-Ⅰ-1,PPb-Ⅰ-1,PPb-Ⅰ-2 and PPc-Ⅰ-1 main chain are mainly composed of(1 → 4)glucoside bond,but its glycosidic bond configuration,molar ratio and connection site There are some differences,resulting in duplication of structural units there are differences.5、Polygonum cuspidatum polysaccharide PPa,PPb,PPc had strong inhibitory activity against α-glucosidase,IC50 values were 114.82 μg / m L,75.68 μg / m L and 70.31 μg / m L,respectively.5 homogeneous polysaccharide components of basic α-glucosidase inhibitory activity is weak.It was deduced that polysaccharides from Polygonum cuspidatum could inhibit the activity of α-glucosidase in combination with chromophores and needed further study. |
