| ObjectiveTo investigate the effects of atorvastatin calcium on insulin secretion induced by potassium chloride in INS-1 cell.MethodsAfter 24 h intervention of atorvastatin calcium in different concentrations(0.2μmol/L、2μmol/L、20μmol/L),ELISA was used to detect the changes of insul in secretion induced by high concentration of potassium chloride in the INS-1 cells.ResultsAtorvastatin calcium can inhibit the insulin secretion(KSIS)in different concentrations(0.2μmol/L、2μmol/L、20μmol/L).There was a statistically significant difference between groups(P<0.01).ConclusionIt is suggested that the atorvastatin calcium may inhibit insulin secretion induced by potassium chloride,and its inhibitory effect shows concentration dependence in INS-1 cell.ObjectiveTo investigate the effect of atorvastatin calcium on autophagy of ENS-I cell.MethodsAfter 24h intervention of atorvastatin calcium in different concentrations(0.2pmol/L、2μmol/L、20μmol/L),the expressions of autophagy-associated proteins such as Beclin 1 and LC3 II in INS-1 cells were examined by Western blot an alysis.The expressions of LC3 II were detected by the Western blot in each group intervened for 24h with 20 umol/L atorvastatin calcium and 20 nmol/L a utophagy late inhibitors,Bafilomycin A1.ResultsCompared with control group,the expressions of Beclin 1 and LC3 Ⅱ protein were increased with atorvastatin calciumthe(P<0.05)in different concentrations(0.2μmol/L、2μmol/L、20μmol/L)in the experimental group.The expression of LC3 ⅡI was significantly increased after the intervention of bflamycin Al(P<0.05).ConclusionIt is suggested that atorvastatin calcium has a certain enhancement effect on the autophagy of the INS-1.The details molecular mechanisms need to be further explored. |
