Objective: To explore the role of BPTF/ c-Myc in proliferation and migration of colorectal cancer(CRC)cell line HCT116 cells;to investigate whether atorvastatin calcium inhibits proliferation and migration of HCT116 cells by down-regulating BPTF/ c-Myc.Methods:1.Clinical specimens of CRC and its adjacent tissues were collected,and the expression of BPTF and c-Myc was detected by immunohistochemistry.2.The expression of BPTF was interfered in HCT116 cells,and the effect of BPTF on the proliferation and migration of HCT116 cells was observed by MTT and scratch experiments.3.c-Myc was overexpressed in HCT116 cells,and its effects on the proliferation and migration of HCT116 cells were observed by MTT and scratch experiments.4.HCT116 cells were treated with atorvastatin calcium(50 μM)and c-Myc inhibitor(10058-F4,F4)(100 μM),respectively,and cell viability and migration were detected by MTT,clone colony assay and scratch assay.At the same time,the protein and m RNA expressions of BPTF and c-Myc in each group were detected by Western blot and real-time quantitative gene amplification fluorescence detection system.Results:1.In the collected clinical specimens of colorectal cancer,immunohistochemistry suggested that both BPTF and c-Myc were highly expressed in human colorectal cancer tissues compared with adjacent tissues.2.After knockdown of BPTF in HCT116 cells,cell proliferation and migration were decreased in the knockdown group compared with normal colorectal cancer cells(p <0.05).3.After overexpression of c-Myc in HCT116 cells,cell proliferation and migration were enhanced in the overexpression group compared with normal colorectal cancer cells(p <0.05).4.Atorvastatin calcium and 10058-F4 can inhibit the proliferation and migration of HCT116 cells.When used in combination,they further inhibit the proliferation and migration of HCT116 cells.5.Both atorvastatin calcium and 10058-F4 can inhibit the protein and m RNA expression of c-Myc in HCT116 cells.When used in combination,they further inhibit the protein and m RNA expression of cMyc in HCT116 cells(p <0.05).6.After knocking down the BPTF in HCT116 cells,adding 10058-F4 could further inhibit the expression of c-Myc protein and m RNA in the knockdown group.7.After overexpression of c-Myc in HCT116 cells,the addition of atorvastatin calcium inhibited the protein and m RNA expression of cMyc in the overexpression group.Conclusion:1.In human colorectal cancer tissues,BPTF and c-Myc are highly expressed and promote the proliferation and migration of HCT116 cells.2.Atorvastatin calcium inhibited the proliferation and migration of HCT116 cells by BPTF/ c-Myc. |
