Objective:To evaluate the efficacy of the tetramethylpyrazine-coated balloon inhibit restenosis after PTA in the New Zeland rabbits iliac arteries injury model.Methods:Thirty New-Zeland White rabbits(weighing 3.0 kg,male)were fed with high-fat diet for 7 days.And the athrosclerosis animal model in one side or both sides of iliac arteries was established by balloon injury via femoral artery strain.2 rabbits were sacrificed at 7,14,28 days after operation respectively.The specimens of iliac arteries were stained with hematoxylin eosin,which proved the model was successful.28 days later,the experiment animals were randomly divided into treatment group(tetramethylpyrazine-coated balloon group,TCB)and control group(plain balloon group,PBA);percutaneous transluminal iliac angiography was performed via common carotid artery.Angiograohy images were recorded and analyzed after four weeks follow-up.Then,the rabbits were sacrificed and target iliac artery segments were removed.The sections were observed by hematpxylin-eosin staining,and some parameters were calculated.Statistical analysis was conducted by SPSS 19.0 software,with the test level set at p<0.05.Results:After balloon injury modeling,hematpxylin-eosin staining confirmed the successful establishment of iliac atherosclerotic stenosis model.Meanwhile,the iliac artery stenosis was observed on the 28 days after operation.During the process of modeling,3 rabbits died due to intraoperative bleeding and excessive anesthesia.Then 21 rabbits(unilateral modeling 6,bilateral modeling 15)enter into the balloon dilation treatment period.11 rabbits(unilateral modeling 3,bilateral modeling 8)were successfully treated with TCB,and 10 rabbits(treated 17 iliac arteries)survival;while 10 rabbits(unilateral modeling 3,bilateral modeling 7)were treated with PBA,and 8 rabbits(treated 13 iliac arteries)survival.At the angiographic follow-up after 4 weeks,TCB group showed lager minimal lumen diameter(1.34±0.15mm vs.0.95 ±0.13mm,p<0.01),lesser diameter stenosis rate(18.11 ±8.84%vs.41.97±7.73%,p<0.01).Pathological analysis showed that comparted with the control group,TCB group had lesser area stenosis rate(46.48+8,22%vs.75.24±6.10%,p<0.01).Conclusions:We successfully established the rabbit model of iliac artery stenosis.TCB could effectively inhibit the neointimal hyperplasia and was superior to plain balloon.
