The blackberry formula prescriptions mainly uses blackberry lyophilized powder as the main raw material,which contains a variety of bioactive chemical substances.And procyanidins and ellagic acid are the most common compounds among them.Procyanidins can be absorbed by the intestinal mucosa and have the function of inhibiting pathogenic bacteria in the intestine.Ellagic acid is metabolized by the intestinal flora to produce urolithins.Urolithin compounds can inhibit the pathogenic bacteria through quorum sensing of the intestinal flora to maintain the balance of intestinal flora.The intestinal flora plays a crucial role in host’s nutrition,metabolism,and immune function.Therefore,it is important for the treatment of inflammation in mice with ulcerative colitis associated colorectal cancer to maintain the stability and balance of the intestinal flora.The results show:(1)Effects and mechanism of blackberry formula on ulcerative colitis associated colorectal cancerIn this study,mice with ulcerative colitis associated colorectal cancer induced by azoxymethane(AOM)and sodium dextran sulfate(DSS)were studied.The 6-week old mice were given a single intraperitoneal injection with AOM,and 2% DSS water solution were given in the next week then the mice were given normal drinking and feeding for 14 days(this cycle was repeated 3 times).Using the blackberry formulas on the first day of modeling,the mice were given enteritis,blackberry formulas II and blackberry formulas IV respectively.In order to investigate the effect of the blackberry formulas on the inflammatory treatment of mice with ulcerative colitis associated colorectal cancer through the regulation of mice intestinal flora.The results showed that compared with the model control group,the blackberry formulas had a significant regulation effect on body weight,colon length,and organ index.The average wet weight of colon was 0.48 g in model control group and was0.38 g in blackberry formulas II.the blackberry formulas IV was 0.40 g.In the determination of disease activity index scores,the model control group was 2.0,the blackberry formulas II and the blackberry formulas IV were about 1.5,the wet weight of colon and DAI scores were higher in model control group than the other groups.The results of observation of colorectal tissue sections showed that compared with themodel control group,the degree of lesions and the infiltration of inflammatory cells decreased significantly in the blackberry formulas.The expression levels of IL-1,IL-6,TNF-α,COX-2,and TGF-β inflammatory cytokines in the blackberry formulas had a decreasing trend compared with the model control group,and IL-1,IL-6,COX-2decreased significantly.According to the above research results,it can be concluded that the blackberry formulas has a therapeutic effect on inflammation in mice with ulcerative colitis associated colorectal cancer.Among them,the blackberry formulas II had a better inhibition on inflammation in ulcerative colitis associated colorectal cancer mice.(2)RT-qPCR detection of changes in flora in colorectal contentsThe content of the probiotics Bacteroides fragilis,Lactobacillus acidophilus,and Eubacterium rectale in the blackberry formulas was higher than the model control group in the intestinal flora of mice with ulcerative colitis associated colorectal cancer for 12 consecutive weeks.There was a significant increase in Eubacterium rectale compared with the model control group.Among them,the blackberry formulas II was more effective than the blackberry formulas IV.The results shows that the expression level of the blackberry formulas was lower than that of the model control group,and the three pathogenic bacteria all had a significant decrease by determinatng content of the pathogenic bacteria Campylobacter,Enterococcus spp and Desulfovibriosp.The content of the probiotics Bacteroides fragilis,Lactobacillus acidophilus and Eubacterium rectale was higher in the blackberry formulas than that of the model control group by determinating the intestinal flora of mice with ulcerative colitis associated colorectal cancer in a 16-week-old blackberry formulas.There was a significant increase in Bacteroides fragilis compared with the model control group.By the determinating the content of pathogenic bacteria Campylobacter,Enterococcus spp,Desulfovibriosp,the expression level of the blackberry formulas was lower than that of the model control group.The Enterococcus spp and Desulfovibriosp were decreased significantly.The effect of promoting probiotics and inhibiting pathogenic bacteria was more significant in the blackberry formulas II than the blackberry formulas IV.The above results showed that the blackberry formulas can promote the growth of intestinal probiotics,and inhibit the growth of pathogenic bacteria.Among them,the effect in blackberry formulas II was more obvious than the blackberry formulas IV.(3)Determination of short-chain fatty acids in fermented metabolites ofintestinal flora by HPLCThe content of short-chain fatty acids,lactic acid,acetic acid,propionic acid,and butyric acid in fermented metabolites of intestinal flora of mice treated with blackberry formulas for 12 weeks was detected by HPLC.The blackberry formulas had an increasing trend compared with the model control group,among which lactic acid,acetic acid,and butyric acid increased significantly.The content of short-chain fatty acids,lactic acid,acetic acid,propionic acid and butyric acid in the metabolites of the intestinal flora of mice treated by the blackberry formulation for 16 weeks was detected by HPLC.The blackberry formulas had an increasing trend compared with the model control group,especially in propionic acid and butyric acid.The promotion effect of lactic acid and acetic acid in the blackberry formulas II was better than that of the blackberry formulas IV.This study investigated the effect of the blackberry formulas on the regulation of intestinal flora to reduce inflammation in mice with ulcerative colitis associated colorectal cancer.By down-regulating the expression levels of inflammatory cytokines IL-1,IL-6,COX-2.Promoting the growth of intestinal probiotics,Bacteroides fragilis Eubacterium rectalein,inhibitions the growth of pathogenic bacteria,Desulfovibriosp,Enterococcus spp,Campylobacter.promotes the production of short-chain fatty acids,a metabolic product of the intestinal flora,and effectively regulated the intestinal environment and the balance of the intestinal flora,In order to reduce the incidence of inflammation in mice with ulcerative colitis associated colorectal cancer.
