Hypertension,and its various complications had become dangerous killers for human health.Cotinus Coggygria,a common species of Anacardiaceae.In the folk of our country,someone found it could significantly reduce blood pressure.Therefore,this experiment was to start from the extraction of the plant and the determination of its ingredients,and then to study whether it had anti-hypertensive and against cardiac remodeling effects,and to explore its mechanism.Methods:1.Two kinds of dry powder were obtained by water and alcohol extraction of Cotinus Coggygria,and the yield of them was compared.Then the content of total flavonoids and total polyphenols were determined by ultraviolet-visible spectrophotometry.2.The isolated rat thoracic aorta was preconstrictid by Norepinephrine,and 50、100、150 and 200μg/mL of water or alcohol extract were added to compare the vasodilatation effect,and the following experiments were carried out by the stronger one.The arterial rings were incubated with L-Name(block NO),Indo(block PGI2),CEA(block KCa)and CsCl(block Kir),through the observation of whether the vasodilatation effect was affected,the mechanism was preliminarily deduced.3.The spontaneously hypertensive rats(SHR)were divided into normal group,model group,positive(captopril)group and 3 Cotinus Coggygria(1,0.1 and 0.01g/kg·d)groups.The changes of blood pressure,weight and heart rate were examined for 12 weeks,and the serum biochemical indexes and cardiac remodeling indicators were examined.Results:1.The yield of water extract was 6.2%,while that of alcohol was5.9%.the total flavonoids content in the aqueous extract and alcohol extract was5.40±0.07 mg/L、7.21±0.07 mg/L,and the total polyphenols content was 4.40±0.08mg/L、2.55±0.04 mg/L.2.The diastolic rates of water extract to vascular rings were21.4±2.1%、49.2±3.5%、76.5±4.3%、94.6±5.5%,and that of alcohol were17.1±1.9%、36.4±3.7%、62.6±5.8%、78.0±7.9%,when the concentration of the extract was 50、100、150 and 200μg/mL.The water extract was significantly stronger than the alcohol one(P<0.01).The vasodilatation rates were 19.1±1.9%,40.4±5.3%,66.4±6.5%,84.1±7.8%in the condition of L-name pre-incubation.The vasodilatation of the aqueous extract was 18.2±1.5%,42.9±4.7%,69.1±6.1%,82.4.0±10.5%in the condition of Indo pre-incubation,and the vasodilatation ability decreased significantly(P<0.01);But there was no effect on the vasodilatation rate in the condition of CEA and CsCl pre-incubation.3.Compared with model group,the systolic and diastolic pressure of high and middle dose group decreased significantly(P<0.001),but there was no significant difference in weight and heart rate of the rats.The serum NO content in the high dose group was 28.5±6.2μmol/L,which was higher than the model group(P<0.05),and the high dose of serum MDA content was 7.06±1.05μg/mL,which was significantly lower than the model group(P<0.01);there was no significant difference in the content of serum ALT,AST and Cr among the groups;LVIDd in the high dose group was5.48±0.26 mm,which was higher than that in the model group(P<0.05),and the high dose group IVSd,LVPWd,LVMI were 2.17±0.13 mm、2.24±0.30 mm、2.98±0.39mm respectively,which were higher than that in the model group(P<0.05).The high dose group heart index was 3.29±0.12 mg/g,which was Less than model group(P<0.05),and there was no significant difference in liver index,spleen index and kidney index among each groups.The content of HPY in the cardiac tissue of high dose group was 338.6±45.24μg/g,which was significantly lower than that in the model group(P<0.05).Conclusion:The yield of water extraction was higher than that of alcohol.The content of total polyphenols in the water extract was higher and the total flavonoid content in the alcohol extract was higher.The water extract and alcohol extract could obviously diastolic blood vessel ring,and the effect of water extract was stronger.The diastolic mechanism was related to the endothelial diastolic factor NO and PGI2,and had nothing to do with the KCa and Kir channels.The high and middle dose can significantly reduce the systolic and diastolic pressure of SHR rats,without affecting the weight and heart rate.There was no obvious harm on the liver and kidney function of SHR rats and had antioxidant effect.And the pathological process of myocardial remodeling and collagen proliferation was inhibited in the high dose group.
