Objective:Study the trend of variability of serum LRG1、COX-2 in patients with chronic alcoholism, explore the mechanism of damage in nervous system caused by alcohol in which LRG1 and COX-2 involved. Methods:Selecting twenty-five patients with chronic alcoholism patients, according to different years of drinking alcohol were divided into drinking less than 20 years’group(include 20 years),and drinking more than 20 years’group;fifteen cases in normal control group. The levels of serum LRG1、COX-2 were measured by enzyme-linked immunosorbent assay. In the experiment, the levels of LRG1 and COX-2 in serum were determined by ELISA double antibody sandwich method.SPSS 22.0 software package was used for statistical analysis. The measurement data that did not meet the normal distribution were represented by the median(upper quartile spacing-lower quartile spacing)M(P75-P25).The non-parametric test was used to compare the chronic alcoholism group with the normal control group;the group drinking alcohol for more than 20 years(including 20 years)and the group drinking more than 20 years,P<0.05 was statistically significant.LRG1 and COX-2 levels in serum if can predict in chronic alcoholism analyzed by ROC curve.Results:1.The LRG1 level in patients with chronic alcoholism 1.89(7.59-0.46)ng/mL lower than the control group 47.54(82.61-15.30)ng/ml,there was significant difference(P<0.001);the COX-2 level was 2.66(4.36-1.32)mg/mL lower than the control group 19.00(51.38-3.20)mg/mL,there was significant difference(P<0.01).2.The LRG1 level in patients with chronic alcoholism has no significance difference(P>0.05)between the two groups:Drinking years ≤ 20years 1.89(3.57-0.82)ng/mL and Drinking years>20years 2.53(3.84-1.45)ng/mL;The COX-2 level in patients with chronic alcoholism has no significance difference(P>0.05)between the two groups:Drinking years ≤ 20years 2.53(3.84-1.45)mg/mL and Drinking years>20 years 2.78(10.03-1.31)mg/mL.3.The area under the curve of chronic alcoholism with LRG1 levels was 0.894(95%CI:0.058-0.944),and the best diagnostic value was 3.80;the area under the curve of chronic alcoholism with COX-2 was 0.726(95%CI:0.508).-0.944),the best diagnostic medium is 5.13.Conclusion:1.The serum levels of LRG1 and COX-2 in patients with chronic alcoholism are reduced and may be involved in the mechanism of alcohol-induced neurological damage.2.With the increase of drinking years, the level of serum LRG1 and COX-2 was not found to changed regularly.3.The levels of serum LRG1 and COX-2 are significant for the prediction of chronic alcoholism.
