| The number of obese people in the world is growing at an alarming rate and obesity seriously threatens human health.And the substances that enter the gut are broken down by the intestinal flora to produce active substance for easy absorption and the complex physiological mechanism under this balance phenomenon is closely related to obesity and other chronic diseases.The anti-obesity mechanism of tea soup and tea powder of different seasons is still not investigated from the perspective of effect on intestinal flora.The regulation of the intestinal flora by the tea soup and tea powder of the black tea of "Fuyun No.6" was studied in rats with high fat diet.The nutritional obesity model of wistar rats was developed by feeding rats with high fat diet.The model rats were gavaged with tea soup and tea powder of spring tea and autumn tea for nine consecutive weeks and obesity-related indicators were analyzed.The result showed that the black tea improved the daily behavior of rats,suppressed the weight gain in rats,reduced the Lee’s index and fat weight,improved the structure of fat cells and increased the number of cells,reduced the content of total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),atherosclerosis index(AI),low density lipoprotein cholesterol(LDL-C),triglyceride(TG)and glucose(GLU)malondialdehyde(MDA),increased the superoxide dismutase(SOD)activity in rat seruum.In addition,the liver damage was alleviated.Using high-throughput sequencing,the intestinal flora were investigated and the result showed that tea soup and tea powder of spring tea and autumn tea could regulate the intestinal flora of rats with high fat diet.The species diversity of intestinal flora were higher in treated groups than in the model group and the number of OTUs varied among groups.The abundance and diversity of intestinal flora were reduced by tea.From perspective of the structure of intestinal flora,the relative abundance of the Firmicutes wasdecreased,the relative abundance of the Bacteroidetes was increased,and the F/B ratio was significantly decreased.In addition,the relative abundance of Ruminococcaceae in the intestinal tract of rats was decreased,and the relative abundance of Lactobacillus,Akkermansia and Lachnospiraceae was recovered by tea.The community structure of intestinal flora in the four groups was similar,and different from the control group.Theaflavins,thearubigins and theabrownine were fed to the high fat diet rat to test their effect on intestinal flora.The result showed that the theaflavins,thearubigins and theabrownine improved the daily behavior of rats,reduced the weight gain and fat mass,alleviated liver injury,and reduced the contents of TC,TG,LDL-C,HDL-C,MDA and AI in serum,increased the content of SOD to suppress the development of obesity.The sequencing data showed that the species diversity of intestinal flora was also higher in the treated groups than in the model group and the number of OTUs in different groups vaired.Theaflavins,Thearubigins and Theabrownine reduced the diversity and abundance of intestinal flora,increased the relative abundance of the Firmicutes and Bacteroidetes,decreased the F/B ratio significantly.In addition,the three components of tea reduced the relative abundance of Ruminococcaceae,increased the relative abundance of Lactobacillus,Akkermansia and Lachnospiraceae.The community structure of intestinal flora was similar among three groups,and different from the control group. |
