Purpose To detect the expression of kinesin family member 20A(KIF20A)in gastric cancer and adjacent normal mucosal tissue,and to explore the relationship between KIF20 A expression and clinicopathological characteristics in gastric cancer(GC).Methods m RNA and protein levels of KIF20 A were detected by quantitative real-time PCR(q RT-PCR)and Western blot in 38 pairs of fresh GC and corresponding normal specimens.After lentivirus-mediated KIF20 A for GC cell lines,cell viability and colony formation assay were carried out to evaluate the effects of KIF20 A on GC in vitro.Immunohistochemistry assay based on a tissue microarray(TMA)consisting of 146 cases was performed to evaluate the prognosis value of KIF20 A.Results We found that KIF20 A expression level is increased in GC(P<0.05).Up-and down-regulating KIF20 A markedly affected cell proliferation in vitro.TMA showed that percentage of KIF20A(+)is significantly higher in GC tissues.Pearson χ2 test method indicated the close relationship between KIF20 A and histological grade(P=0.036).Besides,survival analysis showed that median survival for patients with KIF20A(+)was 40.2 months and for patients with KIF20A(-)was 48.9 months.Therefore,KIF20 A should be an independent prognostic factor.Conclusions 1)KIF20A expression is upregulated at both the transcriptional and translational levels in GC;2)KIF20A promote cell proliferation in vitro;3)Histological grade is poorer in GC patients with KIF20A(+);4)KIF20A is an independent prognostic factor for GC.
