Objective:This study aimed to investigate the expression of KIF20 A in colorectal cancer(CRC)and its correlation with clinicopathologic features and prognosis of CRC patients.To investigate the impact of silencing KIF20 A on the proliferation,invasion and migration of colorectal cancer cells.Methods: Data from TCGA was used to analysis KIF20 A mRNA expression in CRC tissues and adjacent normal tissues.Western blot was performed to examine the protein expression of KIF20 A in our medical center(n=8),and Immunohistochemistry was performed to examine the expression of KIF20 A in 105 CRC tissues.The associations between KIF20 A expression level and clinical data of CRC patients were analyzed by the χ2 tests.Kaplan-Meier survival analysis and log-rank test were carried out to estimate the prognostic significance of KIF20 A expression level in CRC patients.Moreover,univariate and multivariate analysis were used to compare the impact of KIF20 A expression and other prognostic parameters of CRC patients.The expression of KIF20 A of CRC cells was detected by RT-PCR and Western blot.And screened HCT116 and SW1116 which expression of KIF20 A were the highest among the five cells for subsequent cell function experiments.KIF20A-knockdown cells was generated by small interfering RNA,and interference efficiency of KIF20 A was detected by Western blot and RT-PCR.Ability of proliferation,migration and invasion was detected by CCK-8 array,Transwell array and Wound healing assay.Results: 1.KIF20 A was upregulated in the unpaired CRC tissues(n=380)and normal tissues(n=50,P<0.001).The mRNA expression level of KIF20 A in the paired CRC tissues was significantly upregulated than that adjacent normal tissues(n=32,P<0.001).We found that the protein level of KIF20 A was higher in tumor tissues than in normal tissues from our medical center as determined by Western blot(n=8).2.67 patients(64%)were significantly higher expression in CRC tissues,and 38 patients(36%)were lower expression or negative expression(P<0.05).3.Highly expression of KIF20 A in CRC tissues was associated with depth of invasion,lymphatic node metastasis,distant metastasis and TNM stage(P<0.05).Moreover,Kaplan-Meier survival analysis showed that KIF20 A was an independent prognostic factor for both OS and DFS(P<0.001).Cox regression analysis revealed that KIF20 A was an independent prognostic factor in patients with CRC.4.The results of RT-PCR and Western blot showed that HCT116 and SW1116 had a higher expression of KIF20 A,which were chosen for subsequent study.5.CCK-8 array showed that cell proliferative activity was significantly decreased with knockdown of KIF20 A,compared with that in control cells6.Transwell array and Wound healing assay showed that the ability of migration and invasion had decreased after silencing KIF20 A than that of control group.Conclusion: KIF20 A was higher expressed in CRC tissues.Survival analysis showed that KIF20 A high-expression was an independent prognostic factor in CRC patients.The results of this study confirmed that silencing of KIF20 A could inhibit proliferation,migration and invasion capacity of cells.which means that KIF20 A may plays an important role in the tumorigenesis and progression of CRC.
