Regulation Mechanism Of Shugan Jianpi Huoxue Decoction In Notch Pathway Based On Co-Culture Of SEC And HSC

Objective: To observe the effect of Shugan Jianpi Huoxue Decoction(SJHD)serum in the viability of HSC,defenestrae of SEC and Notch pathway by co-culture of SEC-HSC,and study the mechanism of anti-hepatic fibrosis of SJHD.Methods: Extracting drug serum containing SJHD,establishing rat models of hepatic fibrosis,observing histopathology change of livers,the SEC,HSC of normal rats and SEC of fibrotic rats were isolation by collagenase perfusion and Percoll discontinuous density gradient centrifugation.Adding the blocker DAPT,activator rh NF-κB of Notch pathway and SJHD serum in co-culture of SEC and HSC,using MTT method to detect the HSC viability,Western Blot and RT-q PCR methods to detect the CD31,v WF protein expression of SEC and Col Ⅳ,Notch 1,Jagged 1,Hes 1 protein and Notch 1,Jagged 1,Hes 1 m RNA expression of HSC.Results: Compared with normal group,the HSC viability were enhanced in model group(P<0.05,P<0.01).Compared with model group,the HSC viability and Jagged 1 m RNA expression were enhanced in NF group(P<0.05,P<0.01);the HSC viability and CD31,Col Ⅳ,Notch 1,Jagged 1 protein and Notch 1,Jagged 1,Hes 1 m RNA expression were decreased in DAPT group(P<0.05,P<0.01);the HSC viability and Jagged 1 m RNA expression were decreased in SJHD group(P<0.05,P<0.01).Compared with DAPT group,the HSC viability and CD31,v WF,Col Ⅳ,Notch 1,Jagged 1 protein expression were decreased in SJHD combined DAPT group(P<0.05,P<0.01).Compared with NF group,the HSC viability and CD31,v WF,Notch 1,Hes 1 protein and Notch 1,Jagged 1 m RNA expression were decreased in SJHD combined NF group(P<0.05,P<0.01).Conclusion: The mechanism of SJHD on anti-hepatic fibrosis may be related to inhibiting HSC viability,anti-defenestrae of SEC and blocking Notch pathway.
