Objectives:The purpose of this study is to use network pharmacology to predict the mechanism of Zhichan powder in the treatment of Parkinson’s disease.Method:This article adopts the method of network pharmacology to analyze the mechanism of Zhichan powder in the treatment of Parkinson’s disease as follows: through the traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),the active ingredient in Chinese ginseng,Radix Astragali,Magnolia officinalis,Ligustrum lucidum,Paeonia veitchii Lynch were selected by OB ≥ 30%,DL ≥ 0.18,and BBB ≥-0.3,and were selected from the database of TCM in Taiwan.Database@TAIWAN queries the active ingredients of Cynanchum bungei Decne,then supplements the main components of the medicinal materials through literature search,combines the active ingredients of the 7 drugs,and removes duplicates to obtain the effective chemical composition of Zhichan powder.Targets for each component were searched through databases such as TCMSP and BATMAN,and Uniprot database was used to convert target protein names into corresponding gene names.Non-human target proteins and duplicates were removed to obtain target proteins controlled by Zhichan powder.Interacting protein queries and interacting protein network(PPI)constructs were performed using the String database.The GO analysis and the pathway analysis of the interacting proteins were performed by DAVID(National Institute of Allergy and Infectious Diseases),and the action pathways and pathway analysis of Zhichan powder were obtained.Then the Zhichan powder PPI was introduced into the Cytoscape software for network analysis and screening to select the core target of Zhichan powder.Human Parkinson’s disease gene was obtained by NCBI Gene and TTD(Therapeutic Target Database)database.The target gene for Parkinson’s disease was mapped to the Parkinson’s disease target gene.The resulting gene was interrogated by the database of the String database and constructed by interacting protein networks to obtain a network of action of Parkinson’s disease on Parkinson’s disease and explored through pathway analysis.The mechanism of action of Zhichan powder in the treatment of Parkinson’s disease.Result:Network pharmacological analysis results: A total of 52 active Zhichan powders with OB ≥ 30% and DL ≥ 0.18 and BBB ≥-0.3 were obtained(including 18 species of Ginseng ginseng,12 species of Astragalus membranaceus)A total of 22 species and 173 related target proteins were used to construct a protein interaction network of Zhichan powder in the human body through String database.The network analysis was performed using Cytoscape and the core target of Zhichan powder was screened with Degree ≥32,and further analysis of the FDR<0.05 pathway through the enrichment analysis of the DAVID database,found that Zhichan powder can affect molecular functions such as extracellular ligand-gated ion channel activity,GABAergic synapse,chloride transmembrane transport,response to drug,gamma-aminobutyric acid signaling pathway and other biological processes regulate diseases.And it is closely related to the composition of GABA-A receptor complex,postsynaptic membrane,chloride channel complex,and plasma membrane.The DAVID database pathway enrichment analysis revealed that Zhichan powder can have a significant effect on neuroactive ligand-receptor interaction pathways,GABAergic synapses,calcium signaling pathways,dopamine synapses,cholinergic synapses,and p53 signaling pathways,and thus play a role in the treatment of Parkinson’s disease.Through the mapping of Parkinson’s disease gene and atropisia target gene and constructing PPI,the in vivo reaction network of Zhichan powder to Parkinson’s disease patients was obtained,and through DAVID database pathway enrichment analysis,the top 5 pathways were called cell death.Signal pathways of death,p53 signaling,TNF signaling,serotonergic synapses,and neurotrophic factors.In summary,Zhichan powder can treat Parkinson’s disease by acting on immune inflammatory processes and antioxidation.Conclusion:Zhichan powder can be used to treat PD by acting on the process of immune inflammation and antioxidation.
