| Objective To investigate the effects of curcumin on the proliferation,apoptosis and cycle of cultured human keloid fibroblasts(HKF).Methods(1)HKF were cultured in vitro.The HKF were treated with different concentration of curcumin(0,5,10,20 mol/1)on of same generation,and detected cell viability using MTT methods(2)Cell cycle and apoptosis of HKF treated with different concentration of curcumin were detected by flow cytometry.(3)Expression of p27 on the HKF treated with different concentration of curcumin were detected by Western blotResults(1)MTT results showed that culcumin inhibited HKF growth(P<0.01),and the inhibition rate was correlated with culcumin concentration.(2)Using flow cytometry,I found that the apoptosis of HKF was correlated with culcumin concentration(P<0.01).The result also showed that cells in the GO/G1 phase was increased as the increase of drug concentration(P<0.01).(3)Western Blot results showed that p27 was.significantly increased in the concentration dependent manner(P<0.01).Conclusions(1)Culcumin can inhibited HKF growth,and the inhibition rate was correlated with culcumin concentration.(2)curcumin can promote apoptosis of human keloid fibroblasts induced keloid fibroblasts block in GO/G1 phase as the increase of drug concentration,so as to play the role of resistance to scar.(3)The relative expression of p27 protein was significantly increased after the different concentrations of curcumin were used to make the scar tissue.(4)Curcumin can raise the expression of p27,induce the cell block and promotethe cell apoptosis,and ultimately can inhibit the growth of HKF. |
