Objective:To Investigate the regulatory effect of curcumin on proliferation and apoptosis and cell cycle of human cervical carcinoma cell line Hela in vitro. Methods: MTT method was used to assay the antiproliferative effect of curcumin and measure the growth inhibition rates of Hela cells. Cell cycle and apoptosis were inspected by Flow cytometery. Morphological changes of apoptosis cells were observed under light and electron microscopy. In addition, the expression of bcl-2, bcl-xl, caspase-3, cyclinB, p21 and survivin protein in Hela cells were analysed by SP immunohistochemistry. Results: (1)5,10,25,50μmol/L curcumin inhibited the proliferation of Hela cells on a time-and dose-depending manner. (2)The results of Flow cytometric analysis:Hela cells treated with curcumin at 25μmol/L for 24, 48,72 hours were arrested in S and G2/M phase, and induced apoptosis of some cells.(3)The early apoptotic cells were oberved under light microscopy after Hela cells were treated with curcumin at 25μmol/L for 48 hours .At the same condition, Partial cells presented the characteristic morphological changes of apoptosis under the electron microscope. (4)The results of SP immunohistochemistry:The expression of Bcl-2,Bcl-xl and CyclinB was decreased while the expression of Caspase-3 and P21 was increased in Hela cells treated with 25μmol/L curcumin. Conclusion: Curcumin could inhibit the growth of Hela cells; disturbing the cell cycle through up-regulating P21 expression and down-regulating CyclinB and Survivin while inducing apoptosis through up-regulating Caspase-3 and down-regulating expression of Bcl-2, Bcl-xl was probably one of its molecular mechanisms.
