Ochratoxins A(OTA)is harmful to human and animal health,and it is usually found in some moldy grain feeds and products produced by them.The spleen is the most representative immune organ in the body,and it is extremely sensitive to OTA.Astaxanthin(ASX),a member of the carotenoid family,it can participate in regulating the release of cytokines and inhibit cell apoptosis.This experimental study discusses the effect of ASX intervention on OTA-induced mouse spleen toxicity,and explores the molecular mechanism of ASX intervention on OTA-induced mouse spleen toxicity.In this experiment,SPF-grade male C57 mice were randomly divided into 4 groups(Control group,OTA group,ASX group,OTA+ASX group)according to 20 mice in each group.According to the requirements,each group of mice was given a 27-day gavage model,and the spleen and serum of the mice were collected after the experiment.The spleen organ coefficient,spleen pathological section,serum cytokine content,splenic mitochondria-mediated apoptosis pathway related m RNA and protein were detected in each group of mice to study the effect of ASX on OTA-induced spleen toxicity in mice.mechanism.the result shows:(1)OTA reduced the spleen organ coefficient of mice(P<0.01),and damaged the spleen morphology and structure of mice.For example,the boundary between the red and white pulp of the spleen changed from a clear boundary to a blur,the mitochondrial structure was destroyed,and the nucleus Chromatin constriction can also induce a large number of apoptosis of mouse spleen cells and increase the apoptosis rate of mouse spleen cells(P<0.01);ASX has a certain ability to repair the spleen injury of mice induced by OTA,and ASX relieves the small amount of mice induced by OTA.Excessive apoptosis of mouse spleen cells.(2)OTA induces an increase in the levels of IL-6,IL-1β and TNF-α(P<0.01),and interferes with the immune system function;ASX regulates the function of the immune system and inhibits the secretion of IL-1β,IL-6 and TNF-α induced by OTA.(3)In the mitochondrial-mediated apoptosis pathway,OTA down-regulated Bcl-2 m RNA and protein expression in mouse spleen(P<0.01),up-regulated Bax m RNA and protein expression in mouse spleen(P<0.01),and up-regulated mouse spleen The expression of Caspase-3 protein(P<0.01)further promoted the apoptosis of mouse spleen;ASX can interfere with the mitochondrial-mediated apoptosis pathway in mouse spleen and reduce the apoptosis of mouse spleen cells induced by OTA.As far as I am concerned,OTA can induce mouse spleen toxicity;ASX can effectively intervene in the immunotoxicity of mouse spleen caused by OTA.It can regulate the apoptosis of mouse spleen cells by controlling the synthesis of some cytokines and the expression of some apoptosis-related proteins,thereby repairing the spleen damage.
