The SAUR gene(small auxin up RNA)family responds in the early stages of auxin induction and is one of the gene families in the early response of auxin.Creeping bentgrass(Agrostis stolonifera)is a kind of perennial cold season grass of the genus Gramineae,which is widely used in sports fields.SAUR gene function research has only been widely concerned in recent years,but because of the complex gene function of the family,many gene functions have not been clarified,and the gene research is less in the Creeping bentgrass.In the process of genetic transformation,0.3mg/L KT,0.1mg/L 6-BA and 300mg/L were added to callus subculture medium to hydrolyze casein to form embryogenic callus with higher differentiation rate.In the callus induction system,the callus production rate reached 57.2%.During callus induction,the concentration of KT and 6-BA was increased to promote callus differentiation,and finally the callus differentiation rate reached 83.7%.Finally,24 transgenic plants were obtained,7 of which had high expression.The leaves of transgenic plants showed soft curl and premature senescence.The stolon showed that the branching increased and the number of adventitious buds increased significantly.The roots of transgenic plants showed slow growth and decreased complexity.The whole transgenic plants showed slow growth and premature senescence.The IAA content of transgenic plants is generally higher than that of wild type,while the ABA content is relatively low.SAUR71 genes change the morphology and structure of leaves,roots and stolons in different degree,and causes obvious premature senescence in the whole plant.The experimental results show that this gene is important for the regulation of Creeping bentgrass growth and development.SAUR71 gene function has not been studied in previous experiments.SAUR71 gene transformation and preliminary functional analysis were carried out in this experiment,which provided some basis for further exploring gene function.
