| Bovine papillomaviruses(PV)is a neoplastic infectious disease caused by Bovine papillomavirus(BPV).It is characterized by the formation of papillomas on skin and m ucous membranes,and spreads to cattle all over the world.Severe malignant tumor le sions can lead to the death of affected animals and cause certain economic losses to t he cattle industry.According to three aspects to introduce this article:1.Investigation of Bovine Papillomavirus Infection in Southern XinjiangIn this study,samples were collected from many cattle farms,farmers’ markets an d farmers’ households in southern Xinjiang.The suspected disease materials were preli minarily judged through clinical diagnosis,pathological examination,immunohistochemi stry,PCR detection and other methods,and then the suspected disease materials were f urther studied.2.Seroepidemiological survey of bovine papillomavirus in southern XinjiangIn order to understand the infection rate of BPV in cattle in some areas of south ern Xinjiang,the serum of bovine papillomavirus was detected by enzyme-linked react ion kit,and the BPV infection rate in each area was obtained according to the operati on instructions of the kit.3.Molecular Epidemiology of Bovine Papillomavirus in Southern XinjiangIn order to find out the genotype of bovine BPV infection in some areas of sout hern Xinjiang,the universal primer MY11/MY09 and degenerate primer BAPV1/BAPV2 designed according to theconserved sequence of capsid protein L1 gene of papilloma virus have completed the genotyping ofsome epidemic strains in some areas of souther n Xinjiang through sequencing,sequence alignment and genetic evolution analysis.The genotype-specific primer of AKS-02 strain was designed,and the whole geno me sequence of AKS-02 strain was obtained after amplification,sequencing,splicing a nd comparison.The genome of the strain was 7189 bp in length,including 8 open rea ding frames(ORF)of E6,E7,E1,E8,E2,E5,L2 and L1.Phylogenetic tree analysi s showed that AKS-02 strain was BAPV-3 genotype,belonging to Xi genus.The genoty pe-specific primer of AKS-03 strain was designed,and the whole genome sequence of AKS-03 strain was obtained after amplification,sequencing,splicing and comparison.The total genome length of the strain was 7966 bp,including 8 open reading frames(ORF)of E6,E7,E1,E2,E5,E8,L2 and L1.Genetic phylogenetic tree analysis sho wed that AKS-03 strain belongs to BPV-14 genotype,and belongs to Delta genus.This study carried out histopathological examination,PCR detection,immunohistoc hemistry and serological epidemiological investigation on suspected disease materials of bovine papilloma in southern Xinjiang,and genotyped identification of BPV epidemic strains in southern Xinjiang.Two strains of bovine papilloma virus that have been typ ed were selected for full-length genome sequencingand structural feature analysis,provi ding theoretical basis for studying genotyping and sequence structure analysis of bovin e papilloma. |
