Sophora japonica L.has been cultivated for a long time in China.It is an excellent multiple functioning tree species that is the city tree of several cities,has a high landscape value and economic value.In order to keep the good character of mother plant,the techniques of cutting propagation of Sophora japonica were systematically researched,which included cutting media,cuttings parts,times,cuttings roughness,and exogenous hormones,concentration and soaking time.In the meantime,the rooting mechanism was studied by testing dynamic changes of content of endogenous hormones,nutrients and oxidation enzymes activity.Here were the main results:(1)The research of cutting propagation technologyThe results of hardwood cutting display:River sand was the best rooting media.The rooting of cuttings was significantly influenced by the exogenous hormones treatments.The rooting percentage and rooting quality of cuttings treated by NAA were higher than that treated by IBA and ABT1 treatments.Cuttings for 3 hours with NAA 500mg/L resulted in the highest rooting rate,which was up to 78.67%.The rooting quality of cuttings in the middle part was the best.The results of softwood curring display:The mixture substrate of 50%turf and 50%Perlite(v/v)was the best rooting media.The rooting of cuttings was significantly influenced by the exogenous hormones treatments of cuttings.The rooting percentage and rooting quality of cuttings treated by IBA were higher than NAA and ABT1 treatments.Cuttings for 15 seconds with IBA 1500mg/L resulted in the highest rooting rate,which was up to 35.17%.The rooting quality of cuttings in the root sprout was the optimal.The results display:The rooting effectiveness and rooting rate of hardwood cutting is better than that of softwood cutting,moisture in the matrix is the main limiting factor influencing softwood cutting in summer,the high temperature and high humidity can easily cause the cuttings base rot.Callus rooting is the root type of Sophora japonica.(2)The research of rooting mechanismThe results of the nutrient content display:Large amounts of soluble sugar,starch,and protein were consumed.Treatment of exogenous hormones could promote the accumulation of soluble sugar,soluble protein and the hydrolysis and conversion of starch on cutting base,which was beneficial for rooting.The determination results of enzyme activity display:The activities of IAAO,POD and PPO were closely related to rooting of cuttings.Oxidases activities increased during the induction phase and then reduced in different degrees.Preliminary results indicated that exogenous hormones treatments increased enzyme activities during early cutting period and higher activities of IAAO,POD and PPO were beneficial to induction and formation of adventitious root.The test results of endogenous hormones content display:The increase of endogenous IAA content and the ratio of IAA/ABA and IAA/ZR on the base of cuttings could promote adventitious roots induction and formation.But the lower content of ABA,ZR was good to the growth of root.The lower content of GA3 was good for hardwood cutting rooting,while high concentration of GA3 was beneficial to softwood cutting rooting and mainly played an important role in the development of the cuttings of adventitious roots.The test results of rooting-inhibitor display:There were some materials inhibiting rooting of cuttings of Sophora japonica.The contents of these inhibitor materials were different in the different parts of hardwood branches and in different growth periods of softwood,relatively lower in the middle part of hardwood branches and in early July of softwood cuttings.
