Chaenomeles Japonica belongs to Genus papaya, Rose family, also known as Japanese papaya, native to japan.It is loved because its cultivation management is easy and appearance is colorful. cultivation of Chaenomeles Japonica is more in the Shandong,Jiangsu,Zhejiang, Shanxi. linyi of shandong province and suqian of jiangsu province has become important bases of hybrid introduction, test and promotion, cultivated a group of good varieties which produced good economic and social benefits.On the basis of propagation, technology, Chaenomeles Japonica ‘Jyunihitoe’ cutting is a tree of difficult to root at present, mainly through grafting to grow saplings. This study discusses softwood cutting propagation technique of the Chaenomeles Japonica by different cutting media, concentrations of auxin treatment as well as discusses Chaenomeles Japonica softwood cuttage rooting mechanism by cutting roots morphology observation, physiological and biochemical indexes analysis in the process of rooting.The results are as follows:(1) Cuttings rooting of Chaenomeles Japonica ‘Jyunihitoe’ belong to callus rooting types. Cuttings base can grow callus and adventitious roots after hormone treatment.It can significantly improve softwood cuttage rooting effect of Chaenomeles Japonica with the concentration of 225mg·L-1IBA+45mg·L-1 NAA mixed hormone treatment for 60 min.. The rooting rate can reach 82.5%.(2) Different media influence cuttage survival rate of Japanese quince. Compared with sand, vermiculite, peat and vermiculite(1:1), vermiculite and perlite(1:1) mixed medium is the best ideal matrix, callus and adventitious roots appeared earlier, the rooting is significantly better than the other matrix.(3) Rooting related enzyme showed a certain change regulation in a cutting process. The peroxidase(POD) in cuttings increased, then decreased and then increased, while the activity of polyphenol oxidase(PPO) and(IAAO) increased firstly, then decreased, then increased again. Three kinds of enzyme activity and the formation of callus, root and adventitious root formation stages showed obvious corresponding relationship, and effects of auxin treatment on the activity and the trend of the related enzymes were significant.(4) The changes of soluble sugar and soluble protein showed decreased first, then increased, and then decreased trend in a cutting process. Hormone treated cuttings, in the rooting process, C/N ratio in the first decline, and then a little higher, then decreased, and then increased again.In this study, find out softwood cutting rooting mechanism, rich the tree cutting propagation theory; and puts forward the effective softwood cutting propagation technology, provides the technical support for J Chaenomeles Japonica ‘Jyunihitoe’ improved variety breeding.
