C.yunnanensis is a species of cherry blossoms,with good ornamental value.In the study,the adult individual plant YN01,YN02,YN03 were selected as the female parent materials.Study to the rapid propagation system is from the range of obtainment of sterile material,axillary bud induction,proliferation culture,rooting culture and transplanting,etc,in the process of tissue culture,browning and vitrification were studied,and the technology of high efficient tissue culture and rapid propagation was established,which provided the technical support for the realization of large scale of C.yunnanensis.The key findings from this study are as follows:?1?A better procedure for the establishment of sterile lines of C.yunnanensis.The explant should be processed immediately after picking,and the cold preservation time for branches should be within 5 days;the optimal picking time for explant is April,as well as the optimal part,the 4t h to 5th stem cloak counted from the top are collected;following the optimal surface sanitization,1%Bromogeramine eradication for 5 min and 0.1%mercury solution for 28 min,Preservation rate will reach to59%.?2?Induction of axillary buds from C.yunnanensis was screened.The optimum induction medium was MS2+6-BA 1.0 mg/L+N AA 0.1 mg/L+sucrose 30 g/L+agar6.0 g/L,and the highest induction rate was 100%.?3?The culture medium and culture conditions of C.yunnanensis breed culture were optimized.The optimum culture conditions were MS2+6BA1.0 mg/L+N AA 0.05 mg/L+IBA 0.2 mg/L+sucrose 30 g/L+agar 6 g/L.The culture conditions were pH 5.8,light intensity 2000 lx,subculture period 15 days,The temperature is not higher than 23±2?,the maximum proliferation coefficient of up to 4.72,the effective buds rate of about50%,effective buds average of 30 bottles/bottle.?4?Optimized the Rooting C ulture of C.yunnanensis.The optimal medium for rooting was:1/2MS2+NAA 0.5 mg/L+IBA 0.5 mg/L+sucrose 15 g/L+agar 6 g/L,p H 5.8.Rooting time is 10 days,rooting rate>99.3%.?5?Screening out the ideal transplanting substrate of C.yunnanensis.The optimum substrate constitution was peat volume:perlite volume:Horseshoe ash volume=3:1:1,the most suitable transplanting time was from September to November,and the new root grow at the 5th day after transplanting,new leafs came out after the 7th day,transplant survival rate of up to 97%.
