| Estrogens and prostaglandins play an important role in the mammal’s reproductive system.However,there is no report on the mutual regulation between them in dairy cow.Therefore,this study intends to study the effect of estrogen 10-1210-8 mol/L on the prostaglandin synthesis and secretion of PGE2 and PGF2αin dairy cow uterus tissue in vitro.The output of PGE2 and PGF2αwere determined using the competitive enzyme-linked immunosorbent assays.The expression of PGs synthase and PGs receptor genes and proteins in the uterus of dairy cow were detected by transcription-polymerase chain reaction(RT-PCR)and Western Blot,respectively with 10-12 to 10-88 mol/L of estrogen.The results showed that 17β-estradiol could significantly promote the secretion and synthesis of PGE2 in the endometrial tissue of dairy cow in the concentration range of10-10 to 10-8 mol/L(P<0.05),and it was in the range of 10-12 to 10-8 mol/L 17β-estradiol could significantly promote the secretion and synthesis of PGF2αin endometrial tissues of dairy cow(P<0.05).When 17β-estradiol concentration was 10-12 to 10-8 mol/L,the expression of PTGS1,PTGS2,PGES,PGFS,PTGER2,PTGER4,and PTGFR genes and proteins could be upregulated in dairy cow endometrium(P<0.05),significantly down-regulated the expression of CBR1 gene and protein(P<0.05).It can be concluded that estrogen can promote the secretion of PGE2 and PGF2αin the uterus of dairy cow,and inhibit the conversion of PGE2 to PGF2α. |
