| Piriformospora indica is a root-interacting endophytic fungus and has been found in the Indian Thar-Dessert.It colonizes the roots of a wide variety of plant species including monocots,dicots and bryophyte.The Piriformospora indica promotes plant growth,improves plants quality and increases plant resistance and tolerance to stress.Rape(Brassica napus.L),a cruciferous(Cruciferae)canola(Brassica plants),is one of the most important oilseed crops worldwide.This paper uses Liquid chromatography-mass spectrometry(LC-MS)to identify the P.indica-cultured rapes roots proteins.The result shows that:(1)Co-cultured rape overall grew better than control group while single co-cultured plant fresh weight was 2 times as fresh weight control single plant.With Piriformospora indica,rape root fresh weight,srem fresh weight and leaf weight are respectively 1.91,2.48,and 1.75 times of the control group;Stem length,maximum leaf length and maximum leaf width are respectively 1.69,1.44,and 1.46 times of the control group.We got the result that Piriformospora indica promotes the growth of rape,consistently with the previous research results.(2)Use bioinformatics to analysis the total roots proteins.3797 peptides and 1874 proteins are identified in P.indica-cultured rapes roots totally;4338 peptides and 1910 proteins are identified in the control group;2919 peptides and 1813 proteins are identified are shared.61 proteins are only in P.indica-cultured rapes roots while 97 proteins are only in control group.(3)Use GO(Gene Ontology)to analysis the roots protein function from the biological processes,molecular function and cell component.For biological processes,proteins are mainly about biological processes,cellular processes and protein metabolism,more than 50%.For molecular function,binding,catalytic activity and molecular function are the main functions.For cell components,the proteins are distributed in the cells of different position.Most proteins locate in the cell,cell part and the cell component,others mainly locate the cytoplasm,cell organelles,polymer complexes,cell membrane and organelles.(4)Selected more than 2 times and p<0.05 for differential proteins,got 31 different proteins with 16 up-regulated and 15 down-regulated.Serine hydroxymethyl transferase,peroxidase,carboxypeptidase and protein disulfide-isomerase were up-regulated.We speculated that after cocultivation,host plants produce antioxidant mechanism,at the same time Piriformospora indica regulates rape root glutamic acid metabolism,carboxypeptidase and protein disulfide-isomerase expression to accelerate the growth of rape. |
