The root endophytic fungus Piriformospora indica (Sebacinacea) forms mutualistic symbioses with a broad range of host plants. This fungus promotes plant growth, increases resistance of colonized plants against fungal pathogens and tolerance to abiotic stress. Tobacco (Nicotiana tobacum L.) is one of the most important economic crops, belongs to Solanaceae Nicotiana. The crop is seriously affected by various biotic and abiotic stresses. At present,it mainly adopt chemical control methods for prevention and control of pests and diseases, the chemical method bring the resistance, pesticide residues and environmental pollution problems have become more serious, therefore, we need to find a safe and effective method for controlling tobacco pests imminent. Previous studies have shown that Piriformospora indica-colonized tobacco can improve tobacco bioltic and abiotic stress resistant.This paper established a Pi.indica-tobacco co-culture system, we take the way of non-gel separation techniques, mass spectrometry technology and use high-performance liquid chromatography, mass spectrometry and other equipment, to research the symbiont tobacco proteomics. By differential proteomics research methods and bioinformatics, we find differential proteins between tobacco with Pi. indica and tobacco without Pi. indica.1, Influence of Piriformospora indica on protein expression in tobacco leavesWe have identified 3239 peptides in the roots of P. indica-colonized tobacco, the total protein identified is 869. The number of peptides in the control of tobacco roots is 1082, the total amount of the protein is 770. There are 753 proteins both in P. indica-colonized tobacco and non-colonized tobacco,116 for P. indica-colonized tobacco in a unique,17 peculiar for the control of tobacco. After quantitative analysis of proteins, we find the number of expression difference more than four times is 207, including 1,3-beta-glucanase, malic enzyme and others which expression abundance are down-regulated in P. indica-colonized tobacco. But some proteins, such as the glutamine synthetase, pyruvate kinase, cytosolic isozyme etc, are up-regulated in P. indica-colonized tobacco. Significant differential proteins are mainly involved in material transport and energy metabolism, signal transduction, amino acid synthesis and stress reactions through analysis (GO analysis and KEGG analysis).2、Influence of Piriformospora indica on protein expression in tobacco rootsWe have identified 3529 peptides in the leafs of P. indica-colonized tobacco, the total protein identified is 912. The number of peptides in the control of tobacco leafs is 2921, the total amount of the protein is 869. There are 847 proteins both in P. indica-colonized tobacco and non-colonized tobacco.65 for P. indica-colonized tobacco in a unique,22 peculiar for the control of tobacco. After quantitative analysis of proteins, we find the number of expression difference more than four times is 83, including chlorophyll a-b binding protein 50, putative quinolinate phosphoribosyltransferasel, auxin-induced protein PCNT115 which expression abundance are down-regulated in P. indica-colonized tobacco. But some proteins, such as ribulose bisphosphate carboxylase large chain 1, light harvesting chlorophyll a/b-binding protein etc, were up-regulated in P. indica-colonized tobacco. Significant differential proteins are mainly involved in photosynthesis, material transport and energy metabolism and stress response through analysis (GO analysis and KEGG analysis).In summary, we propose that Piriformospora indica may increase the level of proteins involved in photosynthesis, material transport and energy metabolism, stress response, amino acid synthesis to enhance plant growth and resistance.
