Research About The Rapid Propagation And Micro-grafting Technology Of ’Hong Yang’ Kiwifruit

"Hong Yang" kiwifruit belongs to the red meat variety of Chinese gooseberry,Traditional way of seeding consumes a long time and more money,It is difficult to meet the demand of market.This experiment use the caulicle with axillary bud as the explant for tisssue culture,to establish the efficient regeneration system.Main results are as fellows:The most suitable germination medium for "Hong Yang" kiwifruit is 1.0mg/L 6-BA+0.1mg/L NAA+MS and The sprouting rate reached 84%.The most suitable medium for proliferation of adventitious bud is 2.0mg/L6-BA+0.2mg/LNAA+MS and the proliferation coefficient was as high as 4.54.The most suitable medium for the propagation of resistant rootstock is 1.5mg/L6-BA+0.2mg/LNAA+MS and the proliferation coefficient was as high as 4.34.According to the results of rooting experiment,the following conclusions can be drawn:The best rooting medium of ’Hong Yang’ Kiwifruit isl/2MS+07mg/L IBA and the rooting rate was 95%.The best rooting medium of resistant rootstock is 1/2MS+0.5mg/L NAA and the rooting rate was 93%.Through the study on micro grafting of "Hong Yang" kiwifruit and resistant rootstock we learn that:The most suitable concentration of sucrose for micro grafting seedlings is 30g/L;The most suitable culture temperature is 24℃;Five days of dark culture was more conducive for the survival of grafted seedlings;Non rooting resistant rootstock regeneration seedlings are more suitable for micro grafting.Stocks should be with leaves at the same time scions should be without leaves and grafting seedlings should be placed upright to cultivate.
