| The corpus luteum plays an important role in maintaining ovarian function and early pregnancy.The development and degradation of the corpus luteum involves a series of angiogenesis and apoptosis processes,and ROS is also one of the important causes of apoptosis.Studies have shown that ROS and a variety of antioxidant enzymes can affect the development and degradation of a variety of animal corpus luteum.However,no one has done any research on pig yellow body in this area.Therefore,the purpose of this experiment is to study the changes of ROS and antioxidant enzymes during the development and degeneration of porcine corpus luteum tissue,and to supplement the theoretical basis of the antioxidant mechanism of porcine corpus luteum and provide new ideas.In this experiment,the corpus luteum as a material,through its morphology,progesterone levels and internal blood vessel ratio after paraffin section,the corpus luteum is accurately divided into the early,middle,late and white body.The luteal internal ROS levels in pigs at different stages were detected by frozen sections and DHE fluorescent staining techniques.The expression levels of Mn-SOD,CAT and GPx antioxidant enzymes were detected by PCR.Each set of tests was repeated 3 times.The results showed that the level of ROS in porcine corpus luteum tissue was significantly higher than that in other stages(p<0.01)in the later period,and significantly higher in the mid-stage than in the early stage and white body(p<0.01).The difference between the early stage and the white body was not significant.The expression of Mn-SOD was significantly higher in the early stage than in the middle stage and later stage(p<0.05),and the white body was significantly higher than the middle stage(p<0.05),and significantly higher than the later stage(p<0.01).The middle stage was significantly higher than the latter stage(p<0.05).There was no significant change in CAT expression at any time.The initial expression level of GPx1 gene was significantly higher than that of the middle stage(p<0.05),and it was significantly higher than that of the later stage(p<0.01).The white body was also significantly higher than that of the middle and late stages. |
