Camellia oleifera is an important cash crop in China,camellia oleifera industry is an superiority and characteristic poverty alleviation industry supported by the state.Camellia oleifera shell is the residue produced in the process of camellia oil processing,but at present,it has not been utilized with high value.There are a lot of bioactive molecules in the shell of camellia oleifera,such as flavonoids,tea seed polysaccharides,etc.Extraction of these bioactive components from the shell of camellia oleifera,it can not only prolong the chain of camellia oleifera industry and improve the economic benefits of camellia oleifera industry,but also provide a new raw drug resource for biomedical industry.This paper took camellia oleifera shell as the research object,It had investigate and optimize the extraction process of bioactive constituents from the shell of camellia oleifera,and studied the antioxidant activity and antimicrobial activity of crude extracts in vitro environment,the separation and purification process of the crude extract was also explored,preliminary evaluated the effect of pure extracts used as natural antioxidants applied in edible oils to extension of shelf life,which provided a theoretical basis for the high value utilization of camellia oleifera shell.1?Optimization of extraction technology of bioactive components from camellia oleifera fruit shell.Choose five different polar reagents,extract effective components from camellia oleifera shell by ultrasound-assisted method,the extraction rate of total flavonoids was taken as an index for evaluation,and get the best solvent.the optimum extraction process of the extractant was obtained by Box-Behnken design.The results show that the optimum solvent is 60%acetone solution;the technique of extraction of total flavonoids from camellia oleifera shell by ultrasound-assisted ethanol is:ultrasonic power:297w;ultrasonic time:42min;ethanol concentration:64%;material liquid ratio:1:21(g/mL);under this condition,the extraction rate of total flavonoids is 5.52%.2?Study on antioxidant and antibacterial activities of crude extract of camellia oleifera shell in vitro.To inspect the scavenging effect of camellia oleifera shell extract to five common antioxidant systems as DPPH·?·OH?Fe2+chelate?NO2-?total reducing power;the paper studied the Inhibitory ability of camellia oleifera shell extract to escherichia coli,bacillus subtilis and staphylococcus aureus.The results showed that the extract of camellia oleifera fruit shell had antioxidant activity,and its scavenging ability is related to the mass concentration.Their IC50 are respectively 0.07?0.314?2.927?0.856mg/mL,total reducing power is 2.854(Abs?=700),but it is weaker than the same dose of VC and TBHQ,moreover,the active ingredient can adsorb more ferrous ions;camellia oleifera fruit shell extract has bacteriostatic activity,its inhibition ability is related to the species and concentration of microorganisms.the minimum antimicrobial concentration of extract for escherichia coli,bacillus subtilis and staphylococcus aureu of 108 per mL is 2.5mg/mL?2.5mg/mL?1.25mg/mL,It can strongly inhibit the growth of Staphylococcus aureus.3?Separation and purification of crude extract of camellia oleifera shell.The technology of separating and purifying crude extract of camellia oleifera shell by S-8 resin was optimized.The results showed that the optimum process of static adsorption and elution was:adsorption time 5 h,mass concentration of flavonoids in sample solution is 3.9 mg/mL,resolution time is 1.5h and concentration of analyte solution is 60%;the optimum process of dynamic adsorption and elution was:70 ml sample solution with mass concentration of 5.85 mg/mL was sampled at flow rate of 1 mL/min,under these conditions,the purity of flavonoids in crude extract of camellia oleifera shell could be improved by 100%,the yield was close to 90%.4?Evaluation of antioxidant effect of extracts and purified compounds on several edible oils.Using simulation AOM device,and taking acid value and peroxide value of oils as evaluation indexes to investigate it's influence on the quality change of oils,the antioxidant protective effects of extracts and purified crusts of Camellia oleifera on peanut oil,rapeseed oil,walnut oil and camellia seed oil were investigated.The results showed that the extract of the shell of camellia oleifera had a certain anti-lipid oxidation ability and It can inhibit the increase of acid value and peroxide value of oils;the anti-lipid oxidation ability of camellia oleifera shell extract was positively correlated with its dosage;the extract mainly through postpon the arrival time of critical mutation point of acid value and peroxide value and slow down the rising rate of acid value and peroxide value to achieve anti-lipid oxidation effect.
