| The prevalence of osteoporosis increases year by year and has now reached epidemic proportions,China is particularly serious.Existing drug treatment,although effective,but the inevitable side effects.Therefore,the development of security,effective,non-toxic side effects of drugs for osteoporosis has urgent realistic meaning.Animal bone contains rich collagen protein and a variety of mineral elements,is a natural source of high quality protein,but the current of livestock bone utilization rate is low,the lack of depth development.This study will ovine ossein oligopeptide chelate with calcium chloride,through the response surface optimization process,the preparation of peptide chelate calcium.Construction model of osteoporosis by ovariectomized rats to ovariectomy in rats as a model,sham-operated rats were used as controls,17?-estradiol as a positive control,comparing amino acid chelated calcium,collagen peptides and collagen peptide calcium chelate improvement thereof osteoporosis.This paper examines the peptide calcium ratio,pH,collagen peptide concentration,temperature and time on chelation rate,calcium chelate peptide yield,affect chelate calcium content.The optimum by Plackett-Burman and response surface testing to determine the chelating optimum:quality ratio of 2:1,pH 8.10,the temperature is 40?,time was 30min and collagen peptide concentration of 89.28 mg/mL.Peptide chelate of sheep bone collagen peptide chelate and sheep bone collagen peptide calcium chelate under this condition was respectively 34.95%,34.94%,chelate calcium content in the respectively(9.86 ± 0.23)%,(9.05 ± 0.36)%,the chelating rate was 27.01%,26.32%.By SDS-PAGE gel electrophoresis,time-of-flight mass spectrometry,Fourier transform infrared spectroscopy,amino acid composition analysis,scanning electron microscopy of calcium chelating collagen peptide were identified confirmed that collagen peptide and between calcium ion chelate,collagen peptide calcium chelate formation.Vivo study found:the control effect of collagen peptide calcium chelate on the weight of the rats in the control group was significantly better than that in the other groups;collagen peptide calcium chelate can improve cutting womb weight of ovariectomized rats,suggesting that have estrogen like effects;collagen peptide calcium chelate in dose group improve bone mineral density effect best,even higher than that of the estrogen group;each to bone mineral loss prevent collagen peptide calcium chelate>collagen peptide>calcium amino acid chelate;collagen peptide calcium chelate,low dose group and collagen peptide high dose group can make bone hydroxyproline content recovered to the normal level;group can make ovariectomy in rats blood AKP,StrACP,BGP,Ca,P levels returned to sham operated group,especially to collagen peptide calcium chelate group was the best.The high calcium collagen peptide calcium chelate was identified,and it was proved that the collagen peptide calcium chelate was a new type of calcium preparation.Each group had a trend to restore the bone index and blood biochemical index to the normal level,and its effect on the whole collagen peptide calcium chelate(middle dose group)>collagen short peptide group>amino acid chelate group. |
