BackgroundMaca is a South American herbal medicine introduced into China as a new resource food product. There are multiple product developments that are compatible with traditional Chinese medicine. However, this application lacks the theoretical basis of Chinese medicine, and it is necessary to give Maca medicinal properties to introduce it into Chinese medicine. The determination of the potency of traditional Chinese medicine is the key to the introduction of foreign drugs into traditional Chinese medicine. The research group summarized the relevant Chinese and foreign literatures of maca under the guidance of the theories of traditional Chinese medicine and theoretically deduced the medicinal properties of maca.Based on this, the subject uses pharmacological methods to verify the efficacy of Shugan Jiegan,to provide experimental basis for the theoretical hypothesis of the efficacy of Shuganganyu with Macassar, and to introduce traditional Chinese medicine for Maca and to enter traditional Chinese medicine with traditional Chinese medicine. It lays the foundation for clinical research and explores research ideas and methods for the medicinal properties of new Chinese medicine varieties. The research group put forward the research thinking of "Chinese medicine" for foreign medicines:Under the guidance of the theory of traditional Chinese medicine, comprehensive application of the modern research methods of traditional Chinese medicine medicine, through the "document research → theoretical discussion → drug verification → clinical practice" path to study foreign medicine of traditional Chinese medicine Performance provides a scientific basis for its entry into Chinese medicine clinics. The research group has studied the clinical literature of women’s menopausal syndrome and depression by a large number of maca, and theoretically deduces the medicinal properties of maca, and points out that maca can relieve liver qi stagnation,be used for liver qi stagnation, and suffer from chest tightness.Feelings of discomfort, depression and unpleasantness.According to pharmacological experiments, liver qi stagnation model was replicated to observe the effect of maca on liver depression syndrome. It was eventually concluded that the maca is sweet, pungent, and flat;kidneys, liver, and spleen. Can "soothe the liver and dissolve the stagnation",attending liver disorders,qi uncomfortable.Liver and more qi,Shuganjieyu is an important feature of Maca application.Liver stagnation syndrome refers to liver deficiency and catharsis,qi stagnation, and symptoms such as emotional depression, chest pain or low abdominal pain. Also known as liver qi stagnation card. There is a certain correlation between the physiological function of the main hepatic dissociation in Chinese medicine and the regulation of the nervous system on the hypothalamus-pituitary-adrenal(HPA)axis and other body reaction axes and neurotransmitters, and they are involved in the regulation mechanism of the NEI network.Obviously, there are many similarities between the syndrome of liver stagnation syndrome and the regulatory mechanism of NEI.Therefore, taking the physiological and pathological changes of the NEI network in the animal model of liver depression syndrome as the research object, the nature of syndromes of liver depression syndrome can be elucidated, and the liver and catgut blood is responsible for catharsis, which is mainly manifested in the operation of qi and blood circulation.Adjust on.Its mechanism of action is related to endocrine, immune, circulation, neural, digestive and reproductive systems.Based on this,it can be said that the efficacy of Maca is closely related to the NEI network. The results showed that maca can act on the HPA axis and brain monoamine neurotransmitters in the nervous system, indicating that Maca has a significant antidepressant effect.ObjectivesThis study is based on the efficacy of maca "Shugan Jieyu" and the animal model of liver stagnation syndrome as the storting point, systematically combing the characteristics of syndromes of liver and stagnation and a large number of Chinese and foreign literature in Maca,as well as the existing pharmacological effects of maca On the basis of this study,we observed the intervention effect of maca on rat model of liver-qi depression.By examining the relevant physiological and biochemical indexes in order to investigate the effect of maca on "Shugan Jieyu" and its mechanism and Syndrome essence of liver depression.Method:In this study, chronic restraint stress was used to replicate the animal model of liver depression syndrome, taking maca as test substance and traditional Jieyu recipe Daoluoyu as positive control drug.By testing the rats’ body weight and behavior test Consumption experiment, open laboratory test, elevated plus-maze test),as well as the card index, thymus index, adrenal index and serum levels of IL-3,IL-6 and TNF-a in serum and ACT,CORT and hypothalamus CRH level, and the levels of NE,DA,5-HII,E,5-HIAA,HVA and DOPAC in rat cerebral cortex and hippocampus were determined by HPLC-ECD.Result1 literature research1.1 Maca research status at home and abroadBy reading a large number of Chinese and foreign literatures, combined with previous research on the medicinal properties of Maca,we concluded that Maca was sweet, flat, owned kidney, liver and spleen. With Bushenyijing, gluten Zhuanggu,Shuganjianpi effect. The study of Maca belong to liver, liver and Jieyu can be the starting point for the pharmacological effects of Maca.1.2Dialectical theory of syndrome of liver depression and its relationship with neuroendocrine-immune networkThrough reading a large number of literatures,we systematically combed the dialectical theory of traditional Chinese medicine on the syndrome of liver stagnation and the relationship between neuroendocrine-immune network theory and related theories of traditional Chinese medicine. Not only mastered the understanding of traditional Chinese medicine and modern medicine on liver depression syndrome, but also clarified the interrelationship between NIE network and liver depression syndrome, which provided preliminary preparations for the study of syndrome differentiation of liver depression syndrome.2 Experimental research2.1 Maca different extract on mouse tail time, force swimming immobility timeResults:Compared with the blank group, the body weight of the Yue Yao Wan group, the high-dose Maca powder group and the escalated group were significantly increased(P<0.01,P<0.05,P<0.001).Compared with the blank group,Xiaoyao Pill group,Maca original powder high, low dose group, macao alcohol increased, low dose group mice hang tail time were significantly reduced(P<0.001).Compared with the blank group,Xiaoyao Pill bolus group,Makao original powder high, low dose group, maca alcohol increased,low-dose group mice forced swimming time was significantly reduced(P<0.001).2.2 Maca extract different types of chronic restraint stress liver depression rat model of behavior, organ index and inflammatory cytokinesResults:Compared with the model group, the average body weight of Xiaoyao Pills group(P<0.01),maca alcohol increased dose group(P<0.01)and Maca original powder high dose group(P<0.05)(P<0.01).The consumption of cassia in maca alcohol group increased significantly(P<0.01),the increase in maca alcohol group(P<0.01)(P<0.01,P<0.01),maca alcohol increased dose group(P<0.01,P<0.05)Dose group(P<0.05)increased the number of closed arms and the number of open arms in rats of Xiaoyao Pill group(P<0.05)(P<0.05).Compared with the model group, the rats in Xiaoyao Pill group(P<0.05,P<0.05)and the esomepirin increased group(P<0.05,P<0.05)increased the thymus index and adrenal index of the rats in the Xiaoyao Pill group(P<0.001),and the content of IL-6 decreased significantly(P<0.01)The content of IL-3 increased significantly(P<0.01),the content of IL-6 decreased significantly(P<0.05)Content was significantly lower(P<0.05).2.3 different extracts of Maca on chronic stress rat model of liver depression HPA axis and monoamine neurotransmittersResults:Compared with the model group, the CORT content of Xiaoyao Pill group was significantly lower(P<0.01),ACTH content was significantly lower(P<0.01),CRH content was significantly lower(P<0.05)(P<0.01),while the content of ACTH decreased significantly(P<0.01).The CORT content of high-dose Makao original powder significantly decreased(P<0.05)(P<0.01),the content of 5-HT increased significantly(P<0.05),the content of NE increased significantly in macao alcohol-treated group(P<0.01)(P<0.05),the content of DA increased significantly(P<0.01)and the content of 5-HT increased significantly(P<0.05).The content of E in high-dose Makao powder high-dose group increased significantly(P<0.05),DA content increased significantly(P<0.01).ConclusionConclusion 1:The results of experiment 1 show that the antidepressant effect of Maca coffee extract and Maca powder is better than that of Maca juice through different behavioral experiments. Conclusions 2:The results of experiments 2 and 3 show that the alcohol extract of Maca and Maca powder can improve the behavioral changes of rats with liver depression to a certain extent, and the improvement mechanism may be related to the regulation of neurotransmitters and the reduction of stress hormones Secretion, inhibition of HPA axis over-activation,regulation of neuroendocrine system function. To sum up,the present study reveals that Maca can play a role in relieving stagnation of liver and resolving stagnation by regulating the neuro-endocrine-immune(NE)network in an animal model of chronic depression caused by liver-qi stagnation and suggests that "Effect"and the nature of the Syndromes of liver depression Syndrome, the nerve, endocrine, immune system related cytokines, hormones, neurotransmitters as a breakthrough to carry out NEI network research. And on this basis, this study by observing the impact of maca on the animal model of liver stagnation, and further reveal the mechanism of action Macaheluo Jiexiu effect for the clinical application of maca provide a scientific basis, and through the liver depression and Maca Shugan Jieyu effect "evidence" research, not only enriched the basic research of TCM syndromes, but also for the study of traditional Chinese medicine to provide a referencesignificance.
