Objective:To explore the microscopic objective basis and material basis of yang-deficiency constitution and balanced constitution biased constitution classification,by using nuclear magnetic resonance(NMR)based metabonomics.Then to apply the analytica 1 method of Psych and Stats by statistical software R-3.3.2,make correlation analysis between the outcomes of metabonomics and the structure of intestinal flora in former experiment.To verify the hypothesis of "Changes of yang-deficiency constitution’s intestinal flora structure influences its metabolic function,and there exists intestinal flora with feature function intestinal flora related to metabolism".Methods:60 subjects in accord with mild physical and yang-deficiency constitution of physical classification criteria were included in this study,and were divided into 2 groups(30 cases in yang-deficiency constitution group,30 cases in normal constitution group)according to constitution types.Faeces of all the cases would be collected.ⅠH-NMR techniques and multivariate statistical analysis would be used to explore the faeces biomarkers of different physical characteristics,confirming potential biological biomarkers.Finally to find feature function intestinal flora related to typical metabolism biomarkers belonging to yang-deficiency constitution and influence covariation with host metabolism.Results:By comparing metabolite spectral features of faeces from yang-deficiency constitution and balanced constitution,with mild physical as control group,the results showed:1.Metabolomics technology could be used as an objective basis for determining constitution which is feasible.Compared with mild physical,yang-deficiency constitution presented qualitative change in metabolism pathways practically.2.Potential biomarkers of yang-deficiency constitution and balanced constitution were founded;the change tendency in concentration were defined as well.The remarkable potential biomarkers of two different constitutions contained Lactate,(δ1.33-1.35)、Alanine(δ1.46-1.50)、Propionate(δ1.05-1.07)、Acetate(δ1.90-1.93)、Butyrate(δ0.88-0.92,1.56,1.57)、Succinate(82.41)、Propionate/Methionine(δ2.16-2.19)、Glucose/AA(δ3-4)。Compared with the contents of faeces in mild physical group,the contents of Lactate(δ1.33-1.35)、Alanine(δ1.46-1.50)、Propionate(δ1.05-1.07)in yang-deficiency constitution group were higher,while the contents of Acetate(δ1.90-1.93)、Butyrate(80.88-0.92,1.56,1.57)、Succinate(δ2.41)、propionate/Methionine(δ2.16-2.19)、Glucose/AA(83-4)in yang-deficiency constitution group were lower.Relatively obvious potential biomarkers exist in the two different constitutions conclude Ileucine/Leucine/Valine(δ0.95-0.97)、Lysine/Leucine/Arginine(δ1.71-1.79)、Glutamate(82.05,2.06)、Lysine(δ3.01-3.04)、TMA(δ2.94,2.95)、Arginine(δ3,25-3.27)、Glycine(δ3.57)、β-glucose(84.65)、α-glucos e(δ5.24,5.25)。Compared with the contents of faeces in mild physical group,the con tents of Ileucine/Leucine/Valine(δ0.95-0.97)、Lysine/Leucine/Arginine(δ1.71-1.79)、Glutamate(82.05,2.06)、Lysine(δ3.01-3.04)in yang-deficiency constitution group were higher,while the contents of TMA(δ2.94,2.95)、Arginine(δ3.25-3.27)、Glycine(83.57)、β-glucose(δ4.65)、α-glucose(δ5.24,5.25)in yang-deficiency constitution group were lower.3.According to literature review and the relevant databases searching,the differenc es of physiological functions and metabolic pathways of metabolite were explained.It came to a conclusion that biological and biochemical variational aspects have special individual differences of amino acid metabolism,energy metabolism,glucose metabolism,neurotransmitter function of organs etc.existed in yang-deficiency constitution and balanced constitution.lt provided clear and accurate metabolomics supports for the yang-deficiency constitution classification objectively.4.By correlation analysis of multivariate statis tical to explore feature function intestinal flora related typical metabolic makers of yang-deficiency constitution and influence covariation with host metabolism.It turns out that Leuconostoc、Rothia which are higher contents in yang-deficiency constitution related to α-glucose、Butyrate、Lactate、Alanine、Acetate、Glutamate、Propionate/Methionine、Succinate、TMA、Lysine、Arginine、Glucose/AA、Defluviitaleaceae_incertae_sedis、Butyricimonas which are higher contents in yang-deficiency constitution related to a-glucose;Megasphaera are higher contents in balanced constitution related to a-glucose,Lach nospira are higher contents in balanced constitution related to Glucose、α-glucose、β-glucose,Ruminococcus are higher contents in balanced constitution related to Glucose、β-glucose、α-glucose、Butyrate、Lactate、Alanine、Acetate、Glutamate、Propionate/Methionine、Succinate、TMA、Lysine、Arginine、Glucose/AA。These enlightenment have practical significance deeply to be used to explore acknowledge about the correlation between the intestinal flora with various complicated influence factors adequately.Conclusions:1.It came to a conclusion that biological and biochemical variational aspects have special individual differences of amino acid metabolism,energy metabolism,glucose metabolism,neurotransmitter function of organs etc.existed in yang-deficiency constitution and balanced constitution.It provided clear and accurate metabolomics supports for the yang-deficiency constitution classification objectively.There were differences between two biased constitutions NMR metabolic profiles,compared with the balanced constitution,the content of biomarkers are lower is the comprehensive traits of metabolites in yang-deficiency constitution typically,certain biomarkers appear reactive increase slightly.Especially the disorders of energy metabolism,glucose metabolism and amino acid metabolism have correlation with descending physiological function of the related viscera.In order to understand molecular foundation about the relationship between intestinal flora and host metabolism,to help improve health of host and diagnose and treat diseases preferably,this experiment found out special metabolic biomarkers by analysis based on metabonomics.2.Acquire feature function intestinal flora related to typical metabolism biomarkers belonging to yang-deficiency constitution and influence covariation with host metabolism,then to verify the hypothesis of "Changes of yang-deficiency constitution’s intestinal flora structure influences its metabolic function,and there exists intestinal flora with feature function intestinal flora related to metabolism"initially.To make a clear that microcosmic mechanism on intestinal flora have
