| [OBJECTIVE]Study the effects of Bo’fa on the recovery of sensory function of the chronic constriction injury(CCI)model rats through the indexes of the behavioral and morphology.Through the expression of proinflammatory cytokine IL-6 and anti-inflammatory factor SOCS3 in serum and spinal cord in exploring the possible mechanism of-Bo’fa in the treatment of neuropathic pain.[METHODS]Use the male SD rats as experimental animal,randomly divided into normal group,sham operation group,model group and Bo’fa group,the chronic constriction injury of the sciatic nerve(chronic constriction,injury,CCI)as model,using the "massage Simulator"(No.:ZL200710187403.1.)operate Bo’fa on rats,select Yinmen(BL37),Chengshan(BL57),Yanglingquan(GB34)orderly with the length of 3 minutes per acupoint and method.1 times a day,10 times each intervention 1 days off,a total of-20 times of intervention,selected 7 days after modeling and intervention after 20 times as two sampling time,by the experiment of the solar-thermal pain threshold to observe the behavior condition of rats.Use the light microscope to observe the morphology of HE staining of sciatic nerve injury,to further explore the morphological evidence of Bo’fa promoting the recovery of Neuropathic pain injury.detect the content of IL-6 and SOCS3in rat serum and spinal cord,and analyse the change of the expression of these indexes,to explore the possible mechanism of Bo’fa in the treatment of neuropathic pain.[RESULTS]1.Results of behavioral:7 days after modeling,the rats in the normal group were normal;rats in the sham operation group limbs mild claudication,claw contracture,pain tolerance and pain tolerance in normal group rats had no significant difference(P>0.05);model group and Bo’fa group rats gradually appear when walking the operative limb claudication status the ankle and foot,pendulous,not flat on the floor,his back foot phenomenon,pain tolerance decreased significantly compared with the normal group(P<0.05),that of sciatic nerve injury,the rats developed hyperalgesia,CCI model was established successfully.Bo’fa intervention after 20 times:the normal group rats were normal disappeared;the rats in the sham operation group a slight limp phenomenon,basically no difference with the action state of the rats in the normal group;no pain model rats sustained remission,postoperative side claw contracture,hanging does not contact with the ground,joint stiffness method;limbs of Bo’fa group are still limping,feet still Don’t put on the ground,but the claw contracture phenomenon has obviously improved compared with the model group,thermal pain tolerance result is still a certain gap with the normal group(P<0.05),but has been significantly reduced compared to model group(P<0.05),indicating Bo’fa can promote the recovery of hyperalgesia,repair abnormal sensation.2.Morphological results of sciatic nerve:After 7 days,the normal group,the sciatic nerve sections of HE rats in the sham operation group was 40 times under light microscope sciatic nerve fibers arranged neatly,closely,axonal myelin structure is clear;the model group and the Bo’fa group was seen myelin scattered,irregular form and more sparse,nerve fiber arrangement is not the rules of interstitial edema.Bo’fa loose intervention after 20 times of normal group,sham operation group,nerve biopsy staining in normal rats,the model group under light microscope axon,myelin degeneration and severe edema,disorganized,unclear structure,and even the emergence of myelinolysis.Bo’fa group showed axonal diameter began to change coarse,thickness of myelin sheath,around axons,although uneven thickness,did not recover to the normal state,but has showed a trend of gradual recovery,improve nerve fibers arranged orderly.Bo’fa can promote the repair of nerve injury.3.Results of ELISA:After 7 days,detected by ELISA,the content of the sham operation group rats serum IL-6 in no significant difference compared with the normal group(P>0.05);model group compared with normal group compared significantly increased(P<0.05),indicating that CCI surgery can promote the inflammatory cytokines production of IL-6 model in the allocation of power.20 after intervention,the expression of IL-6 in rat serum of Bo’fa group compared with normal group had significant difference(P<0.05),but its value decreased significantly,compared with the same group 7 days after modeling,the content about a 30%decline,and gradually close to the normal group;the intervention after 20 times of IL-6 Bo’fa in model rats in group decreased significantly compared with the same period of expression,there was significant difference(P<0.05).The results showed that the method of Bo’fa intervention can down regulate the expression of inflammatory cytokines IL-6 in serum,reduce inflammation,and relieve the pain.4.Results of Immunohistochemical:(1)IL-6 test results:7 days after modeling,the sham operation group rats spinal cord IL-6 immune positive cells scattered,no significant difference with the normal group(P>0.05),model group compared with normal group significantly increased(P<0.05);Bo’fa intervention after 20 times,the expression of IL-6 in rat spinal cord shifting group in the normal group compared with the same period there are still differences(P<0.05),but the same group compared with 7 days after modeling,the expression about a 30%decline;the expression of IL-6 in spinal cord of rats were allocated in approximately the same period of model group 70%,there was significant difference compared with the model group(P<0.05).The expression method of intervention can Bo’fa Ming down-regulation of IL-6 in spinal dorsal horn,reduce inflammation,relieve pain caused by inflammation.(2)SOCS3 test results:7 days after modeling,the expression of SOCS3 in spinal cord of rats in sham operation group compared with normal group,no significant difference(P>0.05).The model group compared with normal group compared with the expression of SOCS3 increased significantly(P<0.05).Bo’fa intervention after 20 times,the rats in the continuous increasing,about 7 days after modeling with groups of expression 1.4 times,there was a significant difference compared with the model group(P<0.05),about the same period model group was 1.3 times the amount,indicating Bo’fa intervention can regulate the expression of SOCS3 to inhibit nerve inflammation and central sensitization and pain relief.5.Results of Western blot:After 7 days,there was no significant difference between the relative content of protein IL-6 in spinal cord of rats in the sham operation and normal group(P>0.05);IL-6 model group compared with normal group relative protein content differences increased(P<0.05).after 20 days of Bo’fa treatment,IL-6 group of spinal cord shifting the rats in the relative content of protein compared with the normal group there was no significant difference,but its content has decreased significantly,and gradually close to the normal level;the relative content of protein IL-6 in spinal cord of rats in the model group in the same period has a certain degree of decline,but the relative content of protein in the same period of shifting method group decreased there is a large difference(P<0.05).[CONCLUSION]1.Bo’fa can improve the thermal and pain tolerance of CCI rats,relieve symptoms of hyperalgesia in CCI rats.2.Bo’fa can promote the regeneration of injured nerve in CCI rats,reducing axonal swelling,and promote the orderly arrangement of axon and nerve fiber.3.Bo’fa can significantly reduce the expression of IL-6 in serum and spinal cord,Inhibition of inflammatory response,relieve neuropathic pain.4.Bo’fa can significantly improve the expression of anti-inflammatory factor SOCS3 in the spinal dorsal horn,Inhibition of inflammatory response,Central sensitization and pain transduction,relieve neuropathic pain.The experimental results show that Bo’fa can significantly improve hyperalgesia,Promote the recovery of nerve cells;Bo’fa can reduce the expression of proinflammatory factor IL-6 in serum and spinal cord,and increase the expression of anti-inflammatory factor SOCS3 in spinal cord,eliminate inflammation,relieve neuropathic pain. |
