| Objective: Topic from the injury of rat serum TNF-αand TNF-βand skelet al muscle cell apoptosis changes of, observation of TNF-α and TNF-β and s keletal muscle cell apoptosis in different phase variation to analyze characteristi cs of TNF-αand TNF-β, skeletal muscle cell apoptosis changes reveal the p athological process of blood stasis in the skeletal muscle cell injurying.Methods: 80 Wistar rats SPF level, weight 200-250 g, male and female in e ach group were randomly divided into 8 groups. Random set control in group C and group(n = 10, the same below), injury after 1 hour group(1H Group),injury after 12 hours group(12H group), injury after 1 day group(1D group),injury after 3 days group(3D group), injury after 5 days(5D group), injury after 7 days(7D group), injury after 14 days group(14D group), the quantitati ve gravity hit play "[1] in rat left thigh muscle injury caused by, and select ratserum and left thigh muscle groups, groups of TNF-α and TNF-β changes were determined by enzyme linked immunosorbent assay(ELISA). TUNEL met hod was used to determine the apoptosis of skeletal muscle cells in each grou p. We applicate of SPSS17.0 statistical software for statistical analysis.Result:1. In skeletal muscle cell apoptosis, 1D group, 3D group, 5D group, 7D gr oup, 14 D group was significantly increased than the C group(P < 0.01). Grou p 3D, group 5D, group 7D and group 14 D were significantly higher than thos e in group 1H(P < 0.01), and group 1D was significantly higher than that in group 1H(P < 0.05). Group 3D, group 5D, group 7D and group 14 D were s ignificantly higher than those in group 12H(P < 0.01), and group 1D was sig nificantly higher than that in group 12H(P < 0.05). The 1D group was signifi cantly higher than the C group(P < 0.01), which was significantly higher than that in the 1H group and the 12 H group(P < 0.05). 7D group than the C gr oup, 1H group, 12 H group was significantly higher(P < 0.01) than the 1D gr oup was significantly higher(P < 0.05).2.In terms of the number of TNF- α, 12 H group was significantly higher than the C group(P < 0.01), and the 1H group was significantly higher than t he C group(P < 0.05). The 1H group was significantly higher than the C group and the 14 D group(P < 0.05). 12 H group than C group, 14 D group was s ignificantly higher(P < 0.01), the 1D group, 5D group, 7D group TNF- α si gnificant increase than in the number of 12 H group(P < 0.05).3. In terms of the number of TNF-β 12 H group, 1D group than the C gr oup was significantly higher(P < 0.01), 1H group than the C group was signi ficantly higher(P < 0.05).4. TNF-αand TNF-β in this study is no correlation(P≥0.05); the ratio o f TNF-α and TNF-β and inverse ratio had no significant difference(P > 0.05).Conclusion:1. The changes of apoptosis of skeletal muscle cells in the pathological pro cess of injury and blood stasis in 24 hours is the most significant.2. The abnormal changes of blood stasis syndrome in rats serum TNF-α i njuries mainly occurred within 24 hours, 12 hours after injury reached a high point; may suggested that injury blood stasis syndrome of skeletal muscle cell apoptosis death receptor channel inducing factor.3. The change of serum TNF-β in the serum of rats with injury and bloo d stasis mainly occurred within 24 hours, which may be an important factor to stimulate the apoptosis of skeletal muscle cells in blood stasis syndrome.4. There was no linear correlation between TNF-α and TNF-β. The ratio of and the ratio of the different phases in the process of injury remained relati vely stable, and the negative results were further scientific exploration. |
