New technologies for continuous monitoring during the process of cellular proliferation,development and death are urgently needed in studying cell cultivation,artificial organ,drug screening and cancer diagnosis.However,the currently popular cell analytical technologies such as,flow cytometry,fluorescent dye staining,seem to be time-consuming chemicals-consuming,operation complicated,and can't be real-time and continuous.Here,a new cell electrical impedance detection chip based on microelectrode array and biochip technologies which could provide continuous and real-time detection of cell response to various stimuli was presented in this thesis.The content of this thesis are listed as follow:Firstly,a new low-cost cell electrical impedance detection chip was designed and fabricated.The microelectrode array and culture wells in the chip were produced separately and bonded together to form a cell impedance detection chip.The process of cellular proliferation,development and death thus could be monitored and analyzed based on detection chip.Secondly,the measurement conditions of detection chip were optimized.Choosing HepG2 as measuring object,Analyzed the influence of measuring voltage,measuring frequency and cell concentration thereby determining the optimal measurement conditions.Thirdly,proved the validity of chip and analyzed the change of cell state based on electrical impedance.Applied the cisplatin/mitoxantrone stimulus and voltage stimulus to adherent cell then acquired the corresponding impedance response,proved the validity of chip via microscope image and MTT assay.Thus proved the continuous and real-time detection ability of this cell electrical impedance detection chip.Fourthly,applied the detection chip on suspension cell The experiment results shown that cell electrical impedance chip and analytical method were effective on suspension cell suspension state judgment and concentration measurement.In summary,the cell electrical impedance chip technology could be applied in monitoring cell growth and stimuli-response for the advantages of no-biomarker,nondestructive,real-time,continuous and microanalysis,also has extensive application prospect in cytobiology,artificial organ and drug development.
