Association Of ERCC1 And XRCC1 Gene Polymorphisms With The Efficacy Of AGC Patients Treated With L-OHP-Based Neoadjuvant Hemotherapy

Objective: To investigate the efficacy of neoadjuvant chemotherapy for advanced gastric cancer(AGC)in patients with ERCC1 Asn118 Asn and XRCC1 Arg399 Gln gene polymorphisms and oxaliplatin(L-OHP)chemotherapy.Methods: From peripheral venous blood cells of AGC patients.SNPs were genotyped for ERCC1 Asn118 Asn and XRCC1 Arg399 Gln polymorphisms using allele-specific PCR combined with DNA sequencing.The therapeutic effect was evaluated by spiral CT.Results:The ERCC1 Asn118 Asn and XRCC1 Arg399 Gln gene polymorphism is related to the new adjuvant effect,and there are differences in the efficacy of chemotherapy between different genotypes.Through data analysis,C / C(ERCC1 Asn118Asn)and G / G(XRCC1 Arg399Gln)efficiencies were 44%(38/86),35%(30/86),C / T + T / T(ERCC1 Asn118Asn)and G / A + A / A(XRCC1 Arg399Gln)21%(18/86)and 19%(16/86)respectively,of which the C / C(ERCC1 Asn118Asn)efficiency was the highest,the C / T + T / T(ERCC1 Asn118Asn)was slightly higher than the G / A + A / A(XRCC1 Arg399Gln)efficiency,C / C(ERCC1 Asn118Asn)group.Conclusion:There is a correlation between the polymorphism of ERCC1 Asn118 Asn and XR-CC1 Arg399 Gln in patients with AGC and the efficacy and sensitivity of NAC in L-OHP.There were differences in the efficacy of chemotherapy between different genotypes.
