| Objective: Recent studies have demonstrated that many of the key roles of omega-3 fatty acids are mediated by two G protein-coupled receptors(GPCRs)in the free fatty acid receptor(FFAR)family,FFAR1(GPR40)and FFAR4(GPR120).mediate,This experimental study is based on clinical research to investigate the expression of FFAR4(GPR120)protein in breast cancer patients and matching normal breast tissue adjacent to breast cancer, and to evaluate its relationship with clinicopathological features, conventional molecular markers of breast cancer and molecular subtype, explore the role of GPR120 in the occurrence and development of breast cancer. Methods: This study collected 125 cases of breast cancer paraffin specimens that were surgically treated in Cancer Hospital of Ningxia Medical University General Hospital from October 2016 to October 2017,and 63 cases of corresponding adjacent normal tissues, which were stained by immunohistochemistry. The detection method was followed by evaluation of the expression of GPR120 protein in breast cancer tissues and adjacent normal breast tissue. Finally, the expression of GPR120 protein in breast cancer was combined with the clinical pathological features of the patients and routinely detected molecular markers, with perform statistical analysis. Results: The results of immunohistochemistry showed that the positive expression rate of GPR120 protein in breast cancer tissues was as high as 90.4%,which was significantly higher than that in the adjacent normal breast tissues(P<0.001);The positive expression rate of GPR120 protein in breast cancer tissues with high Ki67 expression was as high as 95.1%,while the positive expression rate of GPR120 protein in breast cancer patients with low Ki67 expression was 81.4%,and the difference was statistically significant(P<0.05),The positive expression rate of GPR120 protein in breast cancer tissues was as high as 100% in ER-negative breast cancer patients, and positive expression rate was 85.5% in ER-positive breast cancer patients, and the difference was statistically significant(P<0.05),In addition, the data showed that the positive expression rate of GPR120 protein expression in in situ carcinoma, invasive lobular carcinoma, invasive ductal carcinoma, and other types of cancer increased in turn, and were 66.7%,75%,93.5%,and 100%,respectively.The difference was statistically significant(P<0.05),The statistical analysis results of four different breast cancer molecular classifications showed that the positive expression rates of GPR120 protein in Luminal A,Luminal B,HER-2 over-expression,and TNBC were 85.7%,87.5%,100%,and 95.8%,respectively.The positive expression rate of GPR120 in the HER-2 overexpression and TNBC type breast cancer patients was significantly higher than that of the Luminal type, but the difference was not statistically significant statistical significance. Conclusion: The results of immunohistochemistry showed that GPR120 protein may play a promoting role in the development of human breast cancer tissues;GPR120 protein has a statistically significant difference in different pathological types of breast cancer; The expression of GPR120 protein was positively correlated with the expression of Ki-67 in routinely detected molecular markers in breast cancer tissues and negatively correlated with ER expression. In addition,GPR120 protein had no significant difference in different molecular classification of breast cancer. |
