Objective:To investigate the significance of Bcl-9,VEGF and CD44 in Triple-negative breast carcinoma(TNBC)by detecting and comparing the expressions of Bcl-9,VEGF and CD44 in TNBC and non-triple-negative breast carcinoma(n TNBC)and to analyze the correlationship among the Bcl-9,VEGF and CD44.Methods:176 cases of breast cancer samples were used in this study,including 68 cases of TNBC and 108 cases of nTNBC,whose age and weight matched to each other.IHC was used to detect the expressions of Bcl-9,VEGF and CD44 proteins in tumor tissues.Result:1 The expression of Bcl-9 protein was statistically higher in TNBC group than that in nTNBC groupThe results of IHC showed that the positive of Bcl-9 protein was mainly located in nuclei of tumor cell with yellowish to brownish granules.The positive expression ratio were 88.9%(96/108)and 100%(68/68)in nTNBC and TNBC respectively.The Bcl-9 protein expression in TNBC group was statistically higher than that in nTNBC group(P<0.05).2 The expression of VEGF protein was higher in nTNBC group than that in TNBC groupThe results of IHC showed that the positive of VEGF protein was mainly located in cytoplasm of tumor cell with yellowish to brownish granules.The positive expression ratio of VEGF were 61.8%(21/34)and 87%(47/54)in TNBC group and nTNBC group respectively.The expression of VEGF was higher in n TNBC than that in TNBC group(P<0.05).3The expression of CD44 protein decreased in TNBC group compared with that in nTNBC groupThe positive of CD44 protein was mainly located in membrane of tumor cell with yellowish to brownish granules.The positive expression ratio of CD44 were 51.9%(56/108)and 35.3%(24/68)in n TNBC group and TNBC group respectively.The CD44 protein expression in nTNBC group was markly higher than that in TNBC group(P<0.05).4 The comparison of pathological features between triple negative breast cancer and non-triple negative breast cancerIn TNBC patients with ductal carcinoma in situ with early invasion in 4cases,invasive ductal carcinoma in 34 cases,invasive ductal carcinoma in 30 cases,nTNBC in ductal carcinoma in situ with early infiltration in 12 cases,invasive duct 13 cases were grade I of cancer,70 cases of grade II of infiltrating ductal carcinoma and 13 cases of grade III of invasive ductal carcinoma.Twenty-one cases of TNBC with angiocarcinoma and 33 cases of axillary lymph node metastasis were found in nTNBC group 19 cases of vascular tumor suppository,with axillary lymph node metastasis in 23 cases.5The relationship between Bcl-9,VEGF,CD44 and histological grade,lymph node metastasis,tumor thrombus and neural invasion in TNBCOf the 68 TNBC patients,68 were positive for Bcl-9,of which 33 had lymph node metastases,21 had tumor thrombi,and 37 had nerve invasion.In42 cases of VEGF positive expression,lymph node metastasis occurred in 31 cases,vascular tumor thrombus in 21 cases and nerve invasion in 21 cases.24 cases were positive for CD44,of which 24 cases had lymph node metastasis,19 cases had tumor thrombus and 15 cases had nerve invasion.The chi-square analysis showed that the expression of Bcl-9 was not associated with histological grade,lymph node metastasis,tumor thrombus and nerve invasion(χ2 = 0.262,0.002,0.308,0.014,P > 0.05).The expression of VEGF was correlated with histological grade,lymph node metastasis and vascular tumor thrombus(χ2 = 14.214,28.105,16.933,P<0.05).The expression of CD44 was correlated with histological grade,lymph node metastasis and angioma suppository.The difference was statistically significant(χ2 = 16.464,34.266,40.512,P<005).Conclusion:The Bcl-9 expression in TNBC was slightly higher than that in nTNBC group,while the VEGF and CD44 expressions were lower than those in nTNBC group,which suggested that upregulation of Bcl-9 protein plays an important role in pathogenesis of TNBC.The expression of Bcl-9 is not associated with histological grade,lymph node metastases,vascular tumor thrombi and neural invasion in TNBC tissues.VEGF and CD44 are associated with histological grade,lymph node metastases and vascular tumor thrombi in TNBC tissues.
